This $1.5 million cooperative agreement from the Department of Health and Human Services' Food and Drug Administration (FDA) Research program (CFDA 93.103) supports the development and validation of a best practices framework for physiologically-based pharmacokinetic (PBPK) analysis to support model-informed biowaivers of fed bioequivalence studies for Biopharmaceutics Classification System Class II drugs. The University of Florida is the primary awardee. Under the award, the University of Florida will develop an oral PBPK modeling framework to integrate drug and product characteristics with gastrointestinal physiology at the individual and population levels to predict food-formulation and acid-reducing agent (ARA)-formulation interactions. This will help support waiving requirements for fed bioequivalence studies. Model compounds include rivaroxaban, ibuprofen, itraconazole, and ritonavir. Fraunhofer-Gesellschaft and the University of Maryland, Baltimore are subawardees performing in vitro experiments and PBPK modeling to support framework development. The award period is August 1, 2021 through July 31, 2023.
