About this opportunity

The National Center for Advancing Translational Sciences (NCATS), in coordination with other NIH Institutes and Centers, is soliciting applications for research on undruggable target classes within intractable human disease, with an application due date of February 17, 2028. This is a forecast announcement; the formal Notice of Funding Opportunity has not yet been published.

Eligible applicants include 501(c)(3) and non-501(c)(3) nonprofits, public and private higher education institutions, state and county governments, tribal governments and tribal organizations, city and township governments, independent school districts, special district governments, public or Indian housing authorities, small businesses, and for-profit organizations. Non-U.S. entities and foreign components of U.S. organizations are ineligible.

Applicants must identify a human disease-relevant undruggable target class and develop a method or agent selective for one or more targets within that class. "Undruggable" targets include those that cannot be engaged with high affinity through conventional means—small molecules or biologics that reversibly bind as inhibitors, activators, agonists, or antagonists. Examples include intrinsically disordered proteins, RNA, organelles, and metabolites. Applicants must perform preclinical proof-of-concept studies demonstrating modulation of traditionally undruggable targets. Projects should serve as use cases demonstrating modulation of a single undruggable target while also functioning as proof of concept for a larger technology platform or methodology applicable to the undruggable target class as a whole.

The program is classified as an exploratory and early-stage R21 mechanism. Clinical trials are not allowed. The Assistance Listing number is 93.350 (National Center for Advancing Translational Sciences). Award ceilings, award floors, expected number of awards, period of performance, cost share requirements, and evaluation criteria are not yet specified in this forecast document.

Synopsis

The purpose of this Notice of Funding Opportunity (NOFO) is to solicit applications for exploratory and early-stage projects that focus on undruggable target classes within intractable human disease. Applicants will identify a human disease relevant undruggable target class and develop a method or agent that is selective for one or more targets within that target class. This NOFO is intended to jumpstart research that demonstrates innovative strategies to modulate targets that cannot be addressed by established therapeutic types, furthering the development of methods and/or agents selective for undruggable target classes.

Eligibility

Other Eligible Applicants include the following: Alaska Native and Native Hawaiian Serving Institutions; Asian American Native American Pacific Islander Serving Institutions (AANAPISISs); Eligible Agencies of the Federal Government; Faith-based or Community-based Organizations; Hispanic-serving Institutions; Historically Black Colleges and Universities (HBCUs); Indian/Native American Tribal Governments (Other than Federally Recognized); Regional Organizations; Tribally Controlled Colleges and Universities (TCCUs) ; U.S. Territory or Possession; Non-domestic (non-U.S.) Entities (Foreign Organizations) are not eligible to apply.Non-domestic (non-U.S.) components of U.S. Organizations are not eligible to apply.Foreign components, as defined in the NIH Grants Policy Statement, are not allowed.
