About this file

This document is a Justification & Approval (J&A) for a Simplified Acquisition below the Threshold ($10,000 - $250,000) for 10X Genomics Reagent Kits for the National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK). The procurement covers a bundled set of 10X Genomics consumables and an instrument upgrade for single-cell and spatial transcriptomics projects, including Single-cell RNA-seq reagents, TCR-seq kits, Spatial transcriptomics reagents, and an upgrade for an existing Chromium Controller to a Cytassist instrument.

The justification emphasizes that 10X Genomics is the only vendor producing single-cell sequencing reagents using a gel beads in emulsion system on the Illumina platform, with unique features such as single-cell RNA-seq & immune profiling, fixed-cell RNA profiling, and spatial transcriptomics capabilities. Government Scientific Source, Inc. is the authorized distributor identified to provide these supplies through GSA. The document extensively details why alternative vendors cannot be used, including risks to data comparability, research timelines, and the need to maintain technical compatibility with existing datasets. Market research confirmed no compatible alternatives exist that meet the specific research requirements.

Text version

We use the 10X Genomics Chromium Controller instrument to enable our lab to isolate and uniquely tag individual cells, allowing detailed analysis of gene activity in support of our research mission. We require reagent kits that integrate seamlessly with our existing platform because the resulting data must remain fully comparable to the tens of thousands of single-cell profiles we have already generated in order to maintain technical compatibility and data integrity, which are top priorities for this requirement.

In addition:

1. 10x Genomics reagents are engineered exclusively for our current 10X Genomics Chromium controller instrument, with unique barcodes, microfluidic chips, and analysis software.

2. No third-party kits can operate on this platform or produce data compatible with NIDDK’s existing single-cell datasets.

3. We have already generated large single-cell RNA-seq and spatial datasets using 10x reagents in ongoing projects.

4. Switching to alternative vendors/brands of reagent kits would introduce irreconcilable batch effects, forcing us to repeat months of work and compromising comparability.

5. 10x reagent kits come with ISO-certified manufacturing, lot-tracking, and validated protocols that meet NIH standards for rigor and reproducibility.

6. Adapting another chemistry would require new hardware, months of optimization, and retraining, delaying critical milestones and publications.

7. Spatial methodologies by 10X allow for unique customization only available with the 10X platform.

We conducted market research and searches of GSA Advantage, NIH GWAC catalogs, and major scientific suppliers, such as Fisher Scientific, VWR, MilliporeSigma, and we found no compatible alternatives to the 10x platform and found that competing systems, such as Parse Biosciences, lack comparable kits to what we need for our research. We also weighed risks to project timelines, regulatory compliance, and overall resource cost if we attempted to instead switch vendors and re-engineer our current workflows and determined that it is in the lab’s best interest to continue using the current platform and reagent kits.

Therefore, since the 10X Genomics reagent kits have unique features that are essential to our research mission and also in order to align best with NIH reproducibility guidelines, NIDDK’s research mission, and prudent fiscal management, only the 10x Genomics brand of reagent kits will meet our requirements.

C. Describe why only the proposed contractor is capable of providing the requested supply/service

Government Scientific Source (GSS) is an authorized distributor of the required 10X Genomics brand reagent kits and has been identified as a potential source for this requirement because they can provide the supplies through GSA.
