(16) Background: The NIDDK explores how cells maintain homeostasis in various tissues of the digestive, kidney, and related systems and how that balance breaks down to drive disease. To pinpoint the molecular switches that tip systems of the body from healthy to harmful, our institute profiles tissue resident and homing cells (such as inflammatory immune cells) directly within affected tissue.
Standard bulk and microdissection spatial methods average signals across millions of cells and mask the critical differences that spark disease. The 10x Genomics single-cell and spatial kits solve this problem. Single-cell RNA-seq tells NIDDK which genes are turned on, while spatial workflows let us pinpoint where those cells and molecules are pinpointed within tissue. Merging these readouts gives us a highresolution atlas of gene expression across tissues and disease states with healthy controls.
These data are essential for the NIDDK mission: they reveal the exact pathways that malfunction, identify new drug targets, and guide the development of precision therapies to halt or diseases of major body system organs.
(17) Description of the Requirement: The purpose of this procurement is to acquire a bundled set of 10x Genomics consumables and an instrument upgrade needed for single-cell and spatial transcriptomics projects, as follows: Single-cell RNA-seq reagents (Chromium Next GEM v3.1 chemistry), Single-cell RNA + TCR-seq kits for comprehensive immune-repertoire profiling, Spatial transcriptomics reagents for the Xenium In Situ, Visium HD, and Visium HD-3′ workflows, and an instrument trade-up/upgrade of our existing Chromium Controller to a CytAssist. Together, these items will allow high-resolution mapping of cellular states and spatial gene expression across multiple NIDDK projects. See Attachment 1 -Statement of Need, Section 4.0 REQUIREMENTS for brand-name items list.
