About this file
This is a Statement of Work (SOW) for long read sequencing services to support the Global Parkinson's Genetics Program (GP2), which aims to genotype over 150,000 volunteers worldwide to study the genetic architecture of Parkinson's disease. The contractor must perform long read sequencing on 500 human genomic DNA samples using the PacBio Revio platform with HiFi Run-0 (v1) and provide an output of ≥25X per sample.
The 12-month contract requires the contractor to provide DNA fragmentation, size selection, library preparation, and bioinformatics support including variant calling and structural variant detection. The contractor must securely store samples for 3 months after analysis and deliver raw data, UBAM, and VCF files via secure file transport to CARD/GP2's Google Cloud within 6 weeks of initial QC passage. The government will provide the DNA samples with specific molecular weight requirements (>5ug High Molecular Weight DNA with 70% ≥10 kb and 50% ≥30 kb). Work will be performed at the contractor's site with no travel required, and a dedicated project manager must be assigned as the government point of contact.

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs
GENERAL INFORMATION
Title of Project:
Global Parkinson’s Genetics Program (GP2) - Long read sequencing services
Statement of Need and Purpose:
The purpose of this requirement is to procure long read sequencing services for the Global Parkinson’s Genetics Program (GP2) which will research patient samples associated with Parkinson’s Disease.
Background Information and Objective:
The Global Parkinson’s Genetics Program (GP2) is an ambitious program to genotype >150,000 volunteers around the world to further understand the genetic architecture of Parkinson’s disease (PD). There is still much to learn about genetic risk factors and the path to further understanding requires working collaboratively and openly sharing data, processes, and results.
Period of Performance:
12 months after date of purchase award
SCOPE OF WORK
General Requirements:
Independently and not as an agent of the Government, the Contractor shall furnish all the necessary services, qualified personnel, material, equipment, and facilities, not otherwise provided by the Government as needed to perform the Statement of Work below.
Specific Requirements:
Long Read Sequencing (LRS) shall be performed on 500 human genomic DNA samples.
LRS shall be performed on the Revio platform using PacBio Revio DNA HiFi library kits.
LRS shall include a run scale of HiFi Run-0 (v1).
LRS shall include an output of ≥25X per sample (1 sample per SMRT Cell).
Contractor shall provide long read sequencing of samples with >5ug High Molecular
Weight DNA with 70% ≥10 kb (GQN10kb ≥7.0) & 50% ≥30 kb (GQN30kb ≥5.0) as the starting material.
Contractor shall provide customized solutions for DNA fragmentation, size selection, amplifying target genes or regions, and preparing libraries for high molecular weight human DNA samples.
Contractor shall provide optional high molecular weight genomic DNA isolation services on 500 samples using Nanobind extraction kits
Raw data, UBAM, VCF files shall be delivered via secure file transport protocol to CARD/GP2’s Google Cloud
Contractor shall securely store Government samples for 3 months after the analysis is complete.
Contractor shall provide bioinformatics support including variant calling, structural variant detection, expression profiling, denovo assembly and custom analysis solutions.
LEVEL OF EFFORT:
Contractor staff shall provide the assay results secure file transport protocol, along with routine trouble shooting communications.
GOVERNMENT RESPONSIBILITIES
Government will provide 500 human genomic DNA samples, with >5ug High Molecular Weight DNA with 70% ≥10 kb (GQN10kb ≥7.0) & 50% ≥30 kb (GQN30kb ≥5.0) as the starting material to the contractor.
DELIVERY OR DELIVERABLES
Raw data, UBAM, VCF files shall be delivered via secure file transport protocol to CARD/GP2’s Google Cloud 6 weeks after samples pass their initial QC
REPORTING REQUIREMENTS
The contractor is to provide documentation regarding:
a. Long read sequencing
b. High Molecular Weight gDNA isolation
OTHER CONSIDERATIONS
Travel:
The principal place of performance is the contractor’s work site. Travel for contractor employees will not be necessary.
Key Personnel:
Contractor will provide a dedicated project manager to the government as a contact point for the services.
Information System Security Plan:
Not Applicable
Data Rights:
Section 508—Electronic and Information Technology Standards:
The contractor shall comply with Section 508 of the Rehabilitation Act (29 U.S.C. § 794d), as amended by the Workforce Investment Act of 1998 (P.L. 105-220), August 7, 1998.
