About this file
This is a Statement of Work (SOW) for long read sequencing services supporting the Global Parkinson's Genetics Program (GP2), which aims to genotype over 150,000 volunteers worldwide to study Parkinson's disease genetic architecture. The period of performance is 12 months from award date.
The contractor must perform long read sequencing on 500 human genomic DNA samples using the PacBio Revio platform with specific requirements including ≥25X output per sample, processing of High Molecular Weight DNA samples meeting specific quality thresholds (70% ≥10 kb, 50% ≥30 kb), and providing customized DNA fragmentation and library preparation solutions. Key deliverables include raw data, UBAM, and VCF files delivered via secure protocol to CARD/GP2's Google Cloud within 6 weeks of sample QC approval. Additional requirements include daily courier service to NIH Bethesda campus for sample pickup, providing structural variant calling, optional DNA isolation services for 500 samples, and secure sample storage for 3 months post-analysis. The contractor must provide a dedicated project manager as the primary point of contact.

GENERAL INFORMATION
Global Parkinson’s Genetics Program (GP2) - Long read sequencing services
The purpose of this requirement is to procure long read sequencing services for the Global Parkinson’s Genetics Program (GP2) which will research patient samples associated with Parkinson’s Disease.
The Global Parkinson’s Genetics Program (GP2) is an ambitious program to genotype >150,000 volunteers around the world to further understand the genetic architecture of Parkinson’s disease (PD). There is still much to learn about genetic risk factors and the path to further understanding requires working collaboratively and openly sharing data, processes, and results.
12 months after date of purchase award
SCOPE OF WORK
Independently and not as an agent of the Government, the Contractor shall furnish all the necessary services, qualified personnel, material, equipment, and facilities, not otherwise provided by the Government as needed to perform the Statement of Work below.
Long Read Sequencing (LRS) shall be performed on 500 human genomic DNA samples.
LRS shall be performed on the Revio platform using PacBio Revio DNA HiFi library kits.
LRS shall include a run scale of HiFi Run-0 (v1).
LRS shall include an output of ≥25X per sample (1 sample per SMRT Cell).
Contractor shall provide long read sequencing of samples with >5ug High Molecular
Weight DNA with 70% ≥10 kb (GQN10kb ≥7.0) & 50% ≥30 kb (GQN30kb ≥5.0) as the starting material.
Contractor shall provide customized solutions and guidance for DNA fragmentation, size selection, amplifying target genes or regions, and preparing libraries for high molecular weight human DNA samples.
Contractor shall provide optional high molecular weight genomic DNA isolation services on 500 samples using Nanobind extraction kits. Starting material for these optional DNSA isolation services would be any of the following: human whole blood in EDTA, plasm, cultured cells, or brain tissue.
Raw data, UBAM, VCF files shall be delivered via secure file transport protocol to CARD/GP2’s Google Cloud
Contractor shall securely store Government samples for 3 months after the analysis is complete.
