Biological samples collected in the course of this work include plankton (microscopic water organisms) and fish stomach contents (food fish have eaten). Specialized skills and tools are required to process these samples. Working with a qualified contractor is the most cost-effective way for the USGS CAWSC to process samples to ensure that the data are available for decision-makers in a timely manner.c) Definitions/Applicable Documents - Beaver, J.R, Tausz CE, Renicker TR, Holdren GC, Hosler DM, Manis EE, Davidson RM. 2014. The late summer crustacean zooplankton in western USA reservoirs reflects ecoregion, temperature and latitude. Freshwater biology 59:1173-1186.d) Scope - The purpose of this work is to generate management-relevant data that can be used to support the science of the USGS CAWSC and the science needs of decision-makers. This requires the analysis of plankton and fish stomach contents in a manner similar to Beaver et al. 2014.2) WORK REQUIREMENTS a) Technical Requirements. i) Samples collected by the USGS will be processed by the Contractor according to the below requirements:(1) Zooplankton Samples(a) The overall "biovolume" of a sample is to be recorded. This constitutes the volume of a sample consisting of zooplankton and all other material that has settled to the bottom of a sample jar.(b) Zooplankton samples to be processed generally following the methods of Beaver et al. (2014), including any necessary modifications for local taxa. The basic method is generally as follows: Zooplankton samples will be analyzed using the Utermohl technique with a minimum count of 200 organisms. Counts to be made at 100x magnification, and population densities to be estimated as numbers L-1, based on the volume of water sampled. Dry body mass [¿g dry mass (dm)] to be calculated from published length¿mass relationships and overall biomass estimated (¿g dm L-1) based on the volume of water sampled. Ten specimens to be measured for common species but as few as one for rare taxa.(2) Fish Stomach Content Samples(a) Stomach contents of individual juvenile and adult fish are to be processed in a manner generally consistent with the zooplankton samples.(b) The specific individual fish from a sample that are to be processed for gut contents will be determined in consultation with the USGS principal investigator(c) All stomach contents are to be identified to the lowest practical taxonomic level.(d) Individual prey items should be measured, and a volume or weight obtained. Measurements of individual prey items should be made such that the fresh, undigested size of an eaten prey item can be ascertained.(e) All necessary methods should be employed to avoid or minimize categorizing gut contents as "unidentified." (f) In the majority of cases all individual items in a stomach should be processed. In some cases, a subsample may be processed pending consultation with the USGS principal investigator.(g) Dry body mass [¿g dry mass (dm)] of prey items to be calculated from published length¿mass relationships.ii) Quality Control - Specific QA/QC protocols are required to be developed and instituted by the contractor to ensure correct species identification of invertebrates and fish prey.
