About this opportunity
The National Institute on Aging is soliciting R01 applications for discovery of in vivo chemical probes for novel brain targets with an application due date of February 5, 2027, with clinical trials explicitly not allowed. This joint funding opportunity with the National Eye Institute, National Institute on Drug Abuse, and National Institute of Mental Health supports investigators who possess validated chemical hits requiring optimization toward small-molecule probes relevant to aging, drug abuse, mental health, and vision research missions.
Eligible applicants include higher education institutions (public and private), nonprofits (501(c)(3) and non-501(c)(3)), for-profit organizations, state and local governments, Indian tribal governments (federally and non-federally recognized), public or Indian housing authorities, independent school districts, and special district governments. Foreign organizations may apply, though foreign subawards and subcontracts are not permitted.
Applicants must have in-hand starting compounds with validated activity in at least two orthogonal assays demonstrating hundred-fold dose-response ranges, along with established bioassays for testing analog compounds. The NOFO emphasizes discovery of chemical probes—not therapeutic drugs—requiring applications to address biological target novelty, validated hit characterization, iterative assay cascades (in vitro and in vivo), probe criteria definition, critical path timelines, and future advancement plans. Maximum project periods are five years with budgets based on actual project needs. Scientific review evaluates significance and innovation of biological targets, rigor and feasibility of the chemical optimization approach and proposed assays, investigator expertise, and institutional resources. All NIMH-funded applicants must complete a Data Management and Sharing Plan and share assay data, protocols, chemical structures, and synthetic protocols via PubChem; NIMH-funded awardees must additionally share data through the National Institute of Mental Health Data Archive semi-annually. The opportunity is funded under CFDA 93.866 (Aging Research), 93.279 (Drug Use and Addiction Research), 93.867 (Vision Research), and 93.242 (Mental Health Research Grants).
Synopsis
The purpose of this Notice of Funding Opportunity (NOFO) is to support investigators who have the interest and capability to join efforts for the discovery of in vivo chemical probes for novel brain targets. It is expected that applicants will have, in hand, the starting compounds (validated hits) for chemical optimization and bioassays for testing new analog compounds.Through this NOFO, NIH wishes to stimulate research in:Discovery and development of novel, small molecules for their potential use in understanding biological processes relevant to the missions of National Institute of Mental Health (NIMH), National Institute on Drug Abuse (NIDA), National Eye Institute (NEI), and/or National Institute on Aging (NIA) andDiscovery and/or validation of novel, biological targets that will inform studies of brain disease mechanisms.Emphasis will be placed on projects that provide new insight into important disease-related biological targets and biological processes.
Eligibility
Refer to Section III. Eligibility Information in the NOFO for additional information on eligibility.Foreign Organizations/International CollaborationsNon-domestic (non-U.S.) Entities (Foreign Organizations) are eligible to apply.Non-domestic (non-U.S.) components of U.S. Organizations are eligible to apply.Foreign components, as defined in the NIH Grants Policy Statement, are allowed.
