About this opportunity
The Defense Advanced Research Projects Agency (DARPA) is soliciting innovative concepts to design a protein complex capable of synthesizing DNA or RNA sequences directly in living cells using optical signals through a template-free mechanism. The Generative Optogenetics (GO) program seeks to develop a nucleic acid compiler (NAC) that responds to specific light wavelengths to incorporate nucleotide bases, with two primary research objectives: achieving the core capability of in vivo DNA/RNA synthesis via optical signals and developing error mitigation methods to ensure high-fidelity nucleic acid synthesis. The acquisition will proceed through a two-stage process beginning with written abstracts, with selected applicants invited to present an Oral Proposal Package (OPP) at DARPA. Key contract requirements include meeting progressively challenging technical milestones throughout the 42-month program, with Phase 1 lasting 12 months and Phase 2 lasting 30 months, and performers must participate as either a nontraditional defense contractor or nonprofit research institution. The abstract submission deadline is January 16, 2026, with oral presentations estimated to occur approximately 29 days after abstract submission, followed by the overall proposal due date of February 26, 2026. Evaluation will assess technical feasibility, innovation, team expertise, and alignment with program objectives related to nucleic acid synthesis capabilities and error mitigation mechanisms.
This solicitation uses no set-aside designations and involves no named incumbent contractors. DARPA anticipates making multiple awards structured as Other Transaction Prototype (OT-P) agreements, with Phase 1 fixed-price funding of $1.7 million for Research Objective 1 (RO1) alone or $1.99 million for both RO1 and RO2, though the total aggregate funding for the full program is not specified in the solicitation documents. The 42-month program includes a 12-month Phase 1 focused on refining molecular components and a 30-month Phase 2 dedicated to integrating components and demonstrating in vivo functionality, requiring performers to synthesize progressively longer and more accurate nucleic acid sequences. All program information regarding the nucleic acid compiler is designated as Controlled Unclassified Information (CUI) with strict protection protocols, including a 24-month prohibition on public disclosure of NAC sequences following program completion and mandatory DISTAR approval for any publications. Performance will occur at the contractor's facilities, with continuous government involvement and DARPA obtaining access to program results, patents, and data rights; the program includes supplemental support through workshops, pitch events, and engagement with biosecurity, regulatory policy, and commercialization working groups.
Notice text
The Defense Advanced Research Projects Agency (DARPA) is soliciting innovative concepts to design a protein complex that can be expressed in vivo (i.e., in a living cell) and responds to optical signals to synthesize a corresponding DNA or RNA sequence. This technology should enable massless transfer of genetic information to cells expressing the protein complex via a template-free mechanism that incorporates nucleotide bases in response to specific optical signals (e.g., different wavelengths of light). The GO program is focused on two main Research Objectives (ROs): (1) achieving the core capability of synthesizing DNA/RNA directly in living cells using optical signals, and (2) developing error mitigation methods that ensure high-fidelity nucleic acid synthesis. The acquisition process for GO will proceed through a two-stage process, starting with (1) written abstracts which will inform invitations to brief (2) an Oral Proposal Package (OPP) at DARPA.
