About this file

This document is an abstract template for DARPA's Generative Optogenetics (GO) solicitation (DARPA-PS-26-10), providing detailed instructions for proposers seeking to submit research concepts. The opportunity focuses on developing a protein complex capable of synthesizing DNA or RNA sequences in living cells using optical signals, with two primary research objectives: (1) creating a nucleic acid compiler that can synthesize genetic material using light-based signals, and (2) developing error mitigation methods to ensure high-fidelity nucleic acid synthesis.

The abstract submission process is two-staged, beginning with a written abstract not exceeding 5 pages, which, if approved, will result in an invitation to present an Oral Proposal Package (OPP) at DARPA. Proposers must submit a comprehensive document covering an executive summary, technical approach, technology challenges, and team expertise. The solicitation is structured as a Phase 1 Other Transaction Prototype (OT-P) with a 12-month estimated performance period, and proposers are required to provide a cost estimate for both Phase 1 and an anticipated Phase 2. Abstracts must be submitted electronically through the Broad Agency Announcement Tool and will be evaluated for potential further consideration and potential award negotiation.

Abstract Instructions and Template – DELETE THIS PAGE

Use of this template is mandatory for all abstract submissions to the GO Program Solicitation DARPA-PS-26-10. Proposers must utilize Attachment A. This document must include all components described herein and must be submitted in PDF or Microsoft Word formats. All submissions must be written in English, and all pages shall be formatted for printing on 8-1/2 by 11-inch paper with 1-inch margins and font size no smaller than 12-point. Font sizes of 8 or 10-point may be used for figures, tables, and charts.

Abstracts shall not exceed a maximum of 5 pages. Abstracts should be at the UNCLASSIFIED or Controlled Unclassified Information (CUI) level. All proprietary data and information should be appropriately marked.

Technical Papers (Optional, no more than 3)

Proposers are required to submit an abstract before submitting a Oral Presentation Package (OPP). DARPA will respond to abstracts with a statement as to whether DARPA:

a. Invites the proposer submit an OPP and brief the OPP in-person at DARPA or,

b. does not invite the proposer submit an OPP with a rationale for this decision.

An abstract must be submitted prior to any OPP submission. If an OPP is submitted without 1) Submitting an abstract and 2) Receiving an invitation to submit an OPP – that OPP will not be reviewed. Proposers should note that an invitation to submit an OPP does not mean that the proposed research will ultimately be selected for award negotiation. All official notifications will be sent via email to the Technical Point of Contact (POC) and/or Administrative POC identified on the abstract cover sheet.

Abstracts must be submitted per the instructions outlined herein and received by DARPA no later than the due date and time listed in the Overview Information section. Abstracts received after this time and date may not be reviewed. Please visit Proposer Instructions and General Terms and Conditions for instructions on how to submit your abstract through the Broad Agency Announcement Tool.

Executive Summary Describe what is being proposed and what difference it will make (qualitatively and quantitatively) if successful. Describe the innovative aspects of the project in the context of existing capabilities and approaches, clearly delineating the relationship of this work to any other projects from the past and present. Provide a concise overview of how the proposed approach aligns with the GO program’s objectives to develop an optically-addressable, template-free nucleic acid compiler (NAC) capable of functioning in living cells. Recommended length of 1 page.

Proposed Technical Approach Address the following:

· Your technical vision to achieve the goals of this program

· Approach during Phase 1/Plan for Phase 2 tasks

· Overall approach to meet the goals and milestones of Phase 1

· Outlines specific tasks to meet the milestones of Phase 1

· Presentation of, not just reference to, unpublished data that establishes technical feasibility of Phase 1 work Recommended length of approximately 2 pages.

Technology Challenges Address the following:

· Identify specific technical challenges associated with the proposed approach.

· Discuss key risks and challenges to successful development in the GO program and proposed mitigation strategies.

Recommended length of approximately 1.5 pages.

Technical Expertise/Management Plan Provide a brief summary of the expertise of the team, including subawardees and key personnel. While teaming arrangements do not need to be finalized at the time of abstract submission, mention of potential teaming/collaboration arrangements is highly encouraged. Identify a principal investigator for the project and include a description of the team’s organization, including roles and responsibilities. Recommended length of 0.5 page.
