Assay target regions and PCR cycle conditions will not be disclosed until vendor selection due to unpublished government data. Key personnel must hold expertise in environmental DNA laboratory work and bioinformatics analysis, addressing eDNA-specific challenges including reference library limitations and contamination prevention. Deliverables include taxonomic assignments, exact sequence variants, fastq files, and quality control reports, all meeting publication-quality standards and delivered electronically within six weeks of sample intake. The purpose of this amendment is to provide Questions and Answers:

Question 1: Could the PI share the target region info or reference article so we can take a look? If default primers like 16S, ITS, or 18S are acceptable, that's fine; otherwise, we will need the PCR cycle conditions.
Answer 1: The assay is not published yet and so I can¿t share the details until the vendor is selected.

Question 2: The RFQ lists both CLIN 00010 and CLIN 00040 as ¿Metabarcoding Analyses¿ in accordance with Attachment 1. Can you clarify the intended distinction between CLIN 00010 and CLIN 00040, including whether they should be priced separately, whether they represent different assay/sample types, or whether one is an option/additional quantity?
Answer 2: CLIN 00010 and 00040 are for the same type of analyses, just for different funding accounts.
