About this file
This is Amendment 0002 to a Request for Quote (RFQ) for DNA library preparation and metabarcoding sequencing services issued by the U.S. Geological Survey (USGS), Conservation Genetics Lab, Wetlands and Aquatic Research Center in Gainesville, Florida. The solicitation number is 140G0126Q0198, with an effective date of June 25, 2026.
The USGS requires genomic services including molecular library preparations and next-generation sequencing with specialized bioinformatics for an estimated 300 samples divided into multiple batches of 20-50+ samples per batch. The contract is firm fixed-price for a one-year period. Quote submissions must be received by July 2, 2026 at 11:00 AM Eastern Daylight Time, with an anticipated award date of July 16, 2026. Award will be made on a lowest-price technically acceptable basis, considering vendor pricing, technical capabilities, and experience. The acquisition is not a small business set-aside, with NAICS code 541380. All vendors must be registered in the System for Award Management (SAM.gov) to be eligible for contract award. All quotes and correspondence must be emailed to sruggles@usgs.gov. The amendment clarifies technical specifications through a Questions and Answers section, including confirmation that six separate single-target PCR assays per sample will be performed and pooled, and that specific assay details and gene region information will be shared only after vendor selection due to unpublished government data restrictions.

DNA library preparation and metabarcoding sequencing services. This contract supplies genetic sequencing services
Questions & Answers:
Restate Question 1: Could the PI share the target region info or reference article so we can take a look? If default primers like 16S, ITS, or 18S are acceptable, that's fine;
otherwise, we will need the PCR cycle conditions.
Answer 1: The assay is not published yet and so
I cannot share the details until the vendor is selected.
Restate Question 2: The RFQ lists both CLIN
00010 and CLIN 00040 as Metabarcoding Analyses in accordance with Attachment 1. Can you clarify the intended distinction between CLIN
00010 and CLIN 00040, including whether they should be priced separately, whether they represent different assay/sample types, or whether one is an option/additional quantity?
Answer 2: CLIN 00010 and 00040 are for the same type of analyses, just for different funding accounts.
Question 3: Which specific gene regions do you want to target (e.g., full-length 16S for bacteria, ITS for fungi, etc.)? What are the exact custom primer sequences that you want to use for each target? What are the specific PCR protocols (cycling conditions, annealing temperatures, master mix preferences) required for each target?
Answer 3: This is unpublished government data that cannot be shared until BAO approved. The information and will be shared once the vender is selected.
Question 4: Could you clarify if you are running 6 separate single-target PCR assays per sample and pooling them (which is recommended), or attempting a 6-target multiplex PCR in a single tube?
Answer 4: Yes, 6 separate single-target PCR assays per sample and pooling.

Question 5: What is the total number of separate batches you intend to submit, how many individual samples will be included within each specific batch, and when will each batch be sent approximately?
Answer 5: We estimate a total of 300 samples and approximate batch size from 20-50+ per batch.
