Question 1:

Is it correct to assume that the USACE wants the 72 samples spread across one (1) full 10B flow cell dedicated to the project?

Answer (and edit made to Section C.3.c in amendment 0001):

Yes, that is correct.

Question 2:

What would be the approximate weight and dimensions of the package of 72 samples in dry ice for shipment?

Answer:

The 72 samples will be stored in tubes that will fit in a standard 10x10 cryo tube storage box, approximately 6x6 inches and weighing perhaps one pound. However, the weight and dimensions of the entire package including the dry ice should be determined by the contractor- how long are the samples expected to be in transit and what amount of dry ice would be sufficient for that. Note that information on the proposed shipping method is required as part of the offer contents (see page 39).

Question 3:

Can we assume the Lichen biont tissues will be assessed by the metatranscriptomic method. Or is there any other method specified?

Answer (and edit made to Section C.3.b in amendment 0001):

These samples will be pure cultures, not microbial communities, so transcriptopmics (vs. metatranscriptomics) will be appropriate.

Question 4:

What is the data requirement per library in million reads sought? Our average recommendation is up to 50 million reads per sample.

Answer (and edit made to Section C.3.b in amendment 0001):

50 million reads per sample is sufficient.

Question 5:

10B flow cell of NovSeq X is capable of generating 20 billion reads which appears to be too high. Is there any specific reason for this preference?

Answer (and edit made to Section C.3.c in amendment 0001):

The technology was chosen based on previous discussions with sequencing providers, but a lower number of total reads would be acceptable.
