About this file
This document is a Statement of Need (SON) for an Atomic Force Microscope (AFM) Upgrade to a VRS1250 Package. The National Cancer Institute (NCI) is seeking to upgrade its existing AFM capabilities to enable high-speed, high-resolution scanning of chromatin and nucleosomal dynamics in cancer cells.
The key requirements include a VRS scanner that can scan at 45 frames per second with Angstrom resolution, as well as related VRS gas and ambient chambers, probes, and a VRS1250 sample stage. The upgrade must be delivered within 30 days of contract award, include a minimum 12-month warranty, and provide half-day training. Payment will be made in full upon completion and acceptance of all deliverables. The associated federal contract opportunity, Solicitation Number 75N91024Q00149, is a pre-solicitation for the Atomic Force Microscope Upgrade to VRS1250 Package.

STATEMENT OF NEED (SON)
1.0 TITLE
Atomic Force Microscope Upgrade to VRS1250 Package
2.0 BACKGROUND
The CSEM/LRGBE/CCR/NCI’s core mission focuses on understanding chromosome fragility, specifically with how the nucleoprotein complex called chromatin in vivo is mis-regulated. We are also currently investigation whether histone complexes can adopt alternate structural conformations in cancer cells, and the functional consequences of such alterations upon the cancer epigenome.
Understanding these mechanisms will rely heavily on our ability to visualize chromatin and nucleosomal dynamics at almost near real time with an AFM that is capable of scanning at high speeds and yield high frame rates to see changes as they occur.
