About this file
This is a Statement of Need (SON) for the procurement of a high-performance floor ultracentrifuge by the National Cancer Institute's Laboratory of Biochemistry and Molecular Biology. The National Institutes of Health, Office of Logistics and Acquisition Operations is the procuring federal agency. The requirement is for one Beckman Optima XPN 100 ultracentrifuge capable of operating at a maximum speed of 100,000 rpm with programmable acceleration and deceleration profiles to support the laboratory's existing specialized rotors used in Xenopus egg extract preparation. The instrument must be compatible with the laboratory's current experimental workflows and located adjacent to the satellite animal facility to ensure extract quality and reproducibility.
The contractor shall deliver and install the ultracentrifuge with full white-glove service including truck delivery, onsite installation, and basic operator training conducted by a certified Beckman Coulter field service engineer. The contractor must also trade in and remove the existing Beckman Optima XL-100K ultracentrifuge. All deliverables include documentation, manuals, and standard warranty. The replacement equipment is critical to maintaining the laboratory's primary experimental platform for ongoing federally funded cancer research on genome stability and chromosome segregation. This pre-solicitation (Solicitation Number 75N98026Q01019BECKMANCOULTERPCANCI09667) precedes formal solicitation of this requirement.

Statement of Need (SON)
1. Description of Requirement The National Cancer Institute (NCI), Center for Cancer Research (CCR), Laboratory of Biochemistry and Molecular Biology requires the purchase of one high-performance floor ultracentrifuge, including delivery, installation, and operator training, to replace an aging Beckman Optima XL-100K ultracentrifuge. The requirement includes trade-in of the existing instrument.
The required equipment shall provide a maximum operating speed of 100,000 rpm and support the laboratory's existing specialized ultracentrifuge rotors and established experimental workflows used for preparation of Xenopus egg extracts.
2. Purpose and Background The Laboratory of Biochemistry and Molecular Biology uses Xenopus egg extracts as its primary experimental platform to investigate the molecular mechanisms governing genome stability and chromosome segregation. These biological processes are fundamental to understanding cancer development and identifying potential therapeutic targets.
Ultracentrifugation is the critical step in generating high-quality Xenopus egg extracts. The laboratory's experimental program depends upon reliable, reproducible ultracentrifugation performed at the required centrifugal forces using existing specialized rotors. The research cannot proceed without an ultracentrifuge capable of meeting these performance requirements.
3. Required Capabilities The instrument shall:
· Provide a maximum operating speed of 100,000 rpm.
· Support the laboratory's existing specialized ultracentrifuge rotors required for Xenopus egg extract preparation.
· Provide programmable acceleration and deceleration profiles necessary to preserve extract quality and ensure reproducible experimental conditions.
· Be installed and fully operational in the laboratory with manufacturer installation and basic operator training.
· Replace the existing Beckman Optima XL-100K through trade-in.
4. Need for the Requirement The laboratory's research program requires two specialized ultracentrifuge rotors that depend upon the maximum speed capability and programmable acceleration and deceleration profiles provided by this class of instrument. These technical capabilities are essential for generating high-quality Xenopus egg extracts that produce reproducible experimental results.
In addition, the ultracentrifuge must be located adjacent to the laboratory's satellite animal facility. Xenopus eggs must be processed immediately after collection because transporting crushed eggs over longer distances reduces extract quality and negatively affects experimental reproducibility.
Replacement of the current ultracentrifuge is necessary to maintain continuity of the laboratory's primary experimental platform. Without this capability, the laboratory would be unable to produce Xenopus egg extracts required for ongoing studies of genome stability and chromosome segregation, significantly disrupting federally funded cancer research.
5. Deliverables The contractor shall provide:
· One high-performance floor ultracentrifuge meeting the required specifications.
· Delivery, installation, and operational verification.
· Basic operator training by qualified manufacturer personnel.
· Trade-in and removal of the existing Beckman Optima XL-100K ultracentrifuge.
· All documentation, manuals, and standard warranty associated with the equipment.
