The National Heart, Lung, and Blood Institute (NHLBI) Hematology Branch seeks to screen over 200 patient samples with aplastic anemia and bone marrow failure syndromes for low-frequency somatic mutations. The required product must utilize proprietary ArcherDX VariantPlex Anchored Multiplex PCR (AMP) target-enrichment chemistry, incorporate pre-amplification ligation of unique molecular barcodes for error-correcting sequencing, be custom-configured for a specific panel of bone marrow failure and clonal hematopoiesis genes, and be fully compatible with 150 bp paired-end sequencing on Illumina NovaSeq instruments and the laboratory's existing Archer Analysis bioinformatics pipeline. The estimated delivery turnaround is 4-6 weeks after receipt of order. Delivery is required to Building 10, Room 3E-5216, 10 Center Drive, Bethesda, MD 20892, attention Fernanda Gutierrez Rodrigues, PharmD, PhD.
Solicitation number 75N98027Q00027 is being awarded on a non-competitive basis to Integrated DNA Technologies, Inc. (IDT), 1710 Commercial Park, Coralville, IA 52241, as the sole owner and manufacturer of the required proprietary chemistry. NON-COMPETITIVE COMBINED SYNOPSIS / SOLICITATION
Title: DNA Sequencing of Bone Marrow Failure Syndromes (ArcherDX VariantPlex NGS Library Preparation Kits)
(i) This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in FAR Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested, and a separate written solicitation will not be issued.
(ii) The solicitation number is 75N98027Q00027. This solicitation is issued as a Notice of Intent (NOI) for a non-competitive combined synopsis/solicitation to award a contract or purchase order without providing for full and open competition (including brand-name, if applicable).
This acquisition is conducted under the authority of FAR 12.603, Streamlined Solicitation for Commercial Products or Commercial Services, and is not expected to exceed the simplified acquisition threshold. Additional authority includes 41 U.S.C. 1901 and FAR 6.302-3(a)(2)(ii).
The National Institutes of Health (NIH), Office of the Director, Office of Acquisitions, intends to negotiate and award a firm-fixed-price purchase order on a non-competitive basis to Integrated DNA Technologies, Inc. (IDT), 1710 Commercial Park, Coralville, IA 52241, for custom-designed ArcherDX VariantPlex Next-Generation Sequencing (NGS) target-enrichment library preparation kits. Integrated DNA Technologies, Inc. is the sole owner, manufacturer, and distributor of the proprietary ArcherDX VariantPlex Anchored Multiplex PCR (AMP) chemistry, which has been utilized by the NHLBI Hematology Branch since 2019 to perform error-correcting sequencing (ECS) and is required to maintain scientific continuity, prevent technical batch effects across longitudinal patient cohorts, and ensure compatibility with the laboratory’s established Archer Analysis bioinformatics pipeline. All responsible sources may submit a capability statement, proposal, or quotation, which shall be considered by the agency.
(iii) The solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2026-01, effective 03/13/2026.
(iv) The associated NAICS code is 325413 (In-Vitro Diagnostic Substance Manufacturing) and the small business size standard is 1,250 employees. This requirement is not set aside for small business.
(v) Description of Requirement:
The National Heart, Lung, and Blood Institute (NHLBI), Hematology Branch, Hematopoiesis and Bone Marrow Failure Laboratory requires custom-designed ArcherDX VariantPlex Next-Generation Sequencing (NGS) target-enrichment library preparation kits to screen more than 200 patient samples with aplastic anemia and bone marrow failure syndromes (BMF) for low-frequency somatic mutations (single nucleotide variations and indels below 2–4% variant allele frequency).
Salient Characteristics / Technical Requirements:
• Must utilize proprietary ArcherDX VariantPlex Anchored Multiplex PCR (AMP) target-enrichment chemistry to generate high-complexity NGS libraries from low-input DNA.
• Must incorporate pre-amplification ligation of unique molecular barcodes (MBCs) to enable unique molecule counting and error-correcting sequencing (ECS) for accurate distinction of true low-frequency variants from intrinsic NGS sequencing errors.
• Must be custom-configured to target the laboratory’s specific panel of bone marrow failure (BMF) and clonal hematopoiesis genes.
• Must be fully compatible with 150 bp paired-end sequencing on Illumina NovaSeq instruments and directly compatible with the laboratory’s existing Archer Analysis bioinformatics software pipeline established in 2019.
• Estimated Delivery Turnaround Time: 4–6 weeks after receipt of order (ARO).
(vi) Delivery Location:
National Institutes of Health (NIH) / NHLBI Hematopoiesis and Bone Marrow Failure Laboratory, Hematology Branch Attn: Fernanda Gutierrez Rodrigues, PharmD, PhD Building 10, Room 3E-5216 10 Center Drive Bethesda, MD 20892
