(v) Description of Requirement:
The PCS proteomics support at RML relies on a functioning high-resolution/accurate-mass mass spectrometer, a Q Exactive Plus – Thermo Electron North America LLC, to perform analysis on samples from the Laboratory of Neurological Infections and Immunity (LNII), the Laboratory of Virology (LV), and the Laboratory of Bacteriology (LB) in addition to work related to organoids both for initial analysis and in preparation for downstream workflows related to the NIH SOM initiative. This instrument requires an urgent repair with a replacement control board to continue to do this critical work, and due to the difficulties with the samples that are run on this system many projects for DIR investigators can not be performed while it is not operational. The Q Exactive Plus requires OLM parts and installation to maintain adherence on longitudinal studies.
Thermo Electron is the only manufacturer of the Q Exactive Plus mass spectrometer and is the source of the parts required for this repair as well as the service engineers with the experience necessary to repair the instrumentation. The control board needed for the repair in unique to this instrumentation and not available in unused form from other manufacturers or service providers.

STATEMENT OF WORK (SERVICES)
(SOW)
GENERAL INFORMATION
Title of Project:
Parts and Repair of Q-Exactive Plus with Biopharma Option: AMBIS 2284113; PCAIS PCA-NIAID-02428
Statement of Need and Purpose:
This is for the critical procurement of a Quadupole Electronic II unit and the associated travel to install the unit and repair the associated critical instrumentation. These items are for multiple ongoing projects which require the Q Exactive mass spectrometer located within the Rocky Mountain Laboratories for proteomics or peptide analysis, they include ZIAAI001337 looking at tick feeding and pathogen transmission, ZIAAI001339 with Ashley Groshong on the Lyme disease spirochete, Borrelia burgdorferi, ZIAAI000567 with the Hackstadt group on host-parasite interactions, and a host of others. This instrument also works on a variety of organoids and is needed for proteomic work associated with the newly established NIH Standard Organoid Modeling (SOM) center. The instrument being repaired via this order is utilized within NIAID’s Research Technologies Branch for understanding the proteins involved in the pathways of disease, and its location at RML is critical given the difficulties involved in shipping many sample types from the laboratories. This instrument is currently not operational due to the failure of it’s control board.
Background Information and Objective:
The Proteins & Chemistry Section (PCS) provides support for planning and executing proteomics, metabolomics, lipidomics, and bioactive small molecule studies for intramural investigators. This includes assistance with purification, identification, and quantitative analysis using primarily mass spectrometry-based approaches. The section supports all facets of these projects including but not limited to, project design, separation protocols, sample preparation, chemical labeling for quantitative proteomics, derivatization for improved coverage in metabolomics workflows, enrichment for post-translational modification analysis, and post-acquisition data analysis. The section has a presence in both Rockville, MD and Hamilton, MT with the latter providing key support to studies that are not easily shipped due to the nature of the biological systems being studied. In addition, in the fall of 2025 the NIH Standardized Organoid Modeling (SOM) Center was launched with the aim of coordinating existing expertise across the NIH in order to establish best practices and analytical tools for better calibration of organoid models. As part of the NIH SOM Center initiative, the Protein Chemistry section is both expanding its instrumentation and leveraging the existing infrastructure to support this effort.
