About this file
This is a Presolicitation Notice of Intent issued by the National Institutes of Health (NIH) Office of Acquisition and Logistics Management for non-competitive award to a single source. The National Institute of Diabetes and Digestive and Kidney Diseases seeks synthesis scale-up, custom synthesis, purification, and characterization of optimized lead compounds and specialized precursor materials, specifically prodrug analogs and cyclopentane peptide nucleic acid (PNA) precursors. These materials are not commercially available and require specialized synthetic expertise and production capabilities. The synthesized materials will support ongoing preclinical studies to evaluate compound activity, optimize lead candidates, and advance toward future clinical development, with a focus on anti-cancer therapeutics and HIV prevention and treatment agents.
The procurement is classified under NAICS code 325414 with a size standard of $34 million and is conducted under FAR Part 13 Simplified Acquisition Procedures with an estimated value not exceeding $350,000. The intended awardee is CreaGen Inc., located in Woburn, Massachusetts. Responses from interested parties must be received by July 25, 2026 at 9:00 AM Eastern time and should include technical proposals, cost-price proposals, period of performance, Unique Entity ID (UEI), Taxpayer Identification Number (TIN), and business size certification. All respondents must maintain active registration in the System for Award Management (SAM). Responses should be submitted electronically to Mr. Vishnuvajjala Hanumanth Rao, Lead Purchasing Agent, at vishnuh@mail.nih.gov. The government retains discretion to proceed on a non-competitive or competitive basis.

GENERAL INFORMATION
1. Title: synthesis of prodrug analogs and cyclopentane peptide nucleic acid (PNA) precursors
Background Information: The National Institute of Diabetes and Digestive and Kidney Diseases conducts and supports biomedical research to improve the understanding, prevention, and treatment of diseases affecting human health.
Within the Laboratory of Bioorganic Chemistry, the Synthetic Bioactive Molecules Section conducts research focused on the discovery and development of novel therapeutic compounds for the treatment and prevention of cancer and HIV infection. Ongoing studies have identified several promising sulfanylbenzamide-derived compounds and cyclopentane peptide nucleic acid precursor materials with potential applications as anti-cancer therapeutics, HIV prevention and treatment agents, and diagnostic technologies.
To support these research efforts, the Synthetic Bioactive Molecules Section requires synthesis scale-up, custom synthesis, purification, and characterization of optimized lead compounds and specialized precursor materials. These materials are not commercially available and require specialized synthetic expertise and production capabilities to generate quantities sufficient for continued preclinical evaluation.
The materials produced under this requirement will be used in ongoing preclinical studies to evaluate compound activity, optimize lead candidates, and support advancement toward future clinical development.
