About this file

This is a Technical Specification document from the National Oceanic and Atmospheric Administration (NOAA) Great Lakes Environmental Research Laboratory (GLERL) for Microcystin Toxin Analysis. The document provides definitional guidance and technical parameters for chemical standards and reference materials required for laboratory analysis of cyanobacterial toxins in freshwater systems.

The specification defines several key chemical standards and their applications: Laboratory Reagent (LR) grade chemical standards for routine laboratory work and non-critical analysis; Microcystin-RR (MC-RR), a cyclic heptapeptide hepatotoxin used as an analytical reference standard and certified reference material (CRM) for identifying and quantifying algal toxins in environmental water testing; Microcystin-LW, a specific toxic cyanobacterial analog used as a reference material to identify and quantify algal toxin contamination in raw and treated freshwater systems; and [D-Asp3, (E)-Dhb7]-microcystin-RR, an analytical reference material for identifying and quantifying specific microcystin cyanobacterial peptide toxins. The document also clarifies related chemical standards including Low Foam surfactants, Lactoferrin protein standards, Lethal Factor analytical toxin standards, and Microcystin-LY proprietary reference materials. General chemical standards are described as highly purified substances with known, certified concentrations or purity used to calibrate instruments, validate analytical methods, ensure quality control, and maintain regulatory compliance in laboratory settings.

NOAA Great Lakes Environmental Research Laboratory (GLERL) Microcystin Toxin Analysis

Technical Specification

LR (Laboratory Reagent) grade chemical standards are general-purpose reagents with a high purity level suitable for routine laboratory work, educational training, and non-critical qualitative or semi-quantitative analysis, though they lack the ultra-high, certified purity of Analytical Reagent (AR) or primary standard grades.

Chemical standards labeled RR most commonly refer to Microcystin-RR (MC-RR), a cyclic heptapeptide hepatotoxin produced by cyanobacteria (blue-green algae). They are utilized as analytical reference standards and certified reference materials (CRMs) to identify and quantify algal toxins in environmental water testing.

In chemical analysis and industrial processing, "LF" generally stands for Low Foam (as in low-foaming surfactants like Plurafac LF or OxiVantage LF) or Lactoferrin (an LF protein standard used in dairy and biomedical assays). It can also denote Lethal Factor (an analytical toxin standard) or Low-Field NMR calibration standards.

Microcystin-LW is a specific toxic cyanobacterial analog (a cyclic heptapeptide hepatotoxin) produced by blue-green algae. An analytical standard of Microcystin-LW is used in laboratory and environmental settings as a reference material to identify and quantify algal toxin contamination in raw and treated freshwater systems.

Chemical standards (or analytical reference standards) are highly purified substances with a known, certified concentration or purity. Labs use them as a baseline to identify, measure, and test other materials to ensure accuracy, quality control, and regulatory compliance.

Chemical standards in a laboratory (LA) setting are reference materials with precisely known properties or concentrations used to calibrate instruments, validate analytical methods, and quantify unknown chemical substances. They ensure accuracy, reproducibility, and regulatory compliance across testing procedures.

"LY" typically refers to proprietary pharmaceutical research compounds, inhibitors, or specific biochemical reference materials (such as Microcystin-LY) cataloged by manufacturers for laboratory testing, chromatography, and quantitative assay calibration.

The chemical standard [D-Asp3, (E)-Dhb7]-microcystin-RR (often labeled as [D-Asp3, Dhb7]-MC-RR) is an analytical reference material used to identify and quantify specific microcystin cyanobacterial peptide toxins produced during harmful algal blooms.
