Statement of Work for the Super Resolution/Confocal Optical Microscope

1. SCOPE

This Statement of Work (SOW) will be the basis for the procurement of a Super Resolution Microscope (SRM) at Brookhaven National Laboratory. Brookhaven National Laboratory seeks information on an advanced, fully integrated confocal and super-resolution (STED) microscopy system with comprehensive fluorescence lifetime imaging (FLIM/FCS) functionality. The SRM instrument should provide true confocal point scanning with a tunable white light laser excitation source of visible range capable of simultaneous multi-line operation with independent intensity control and rapid microsecond-scale switching between excitation regime, broadband spectral detection in the range approximately 400–850 nm, with multiple independently configurable detection channels, FLIM/FCS functionality, and analytical tools enabling functional parameter mapping and FRET. Super-resolution performance should include both 2D and 3D STED nanoscopy across the visible spectrum, with possibility to acquire super-resolution imaging at ~30 nm lateral resolution. The instrument should operate within a unified software environment supporting automated calibration, lifetime fitting, super-resolution reconstruction, and compatibility with external analysis platforms. This document outlines the Contractor’s responsibilities and obligations necessary for completing the requirements as set forth herein.

1.1 Background

The Center for Functional Nanomaterials (CFN) is a U.S. Department of Energy Nanoscale Science Research Center and User Facility of Brookhaven National Laboratory. CFN staff provides expertise and capabilitiesto the science user community from national laboratories, academia and industry to accelerate the scientific discovery and innovation. The CFN supports various DOE programs through its user support and in house research science program. The installation of a SRM instrumnet strengthens BNL’s capability in studying materials at nanoscale and investigating processed beyond diffraction limited resolution with non destructive optical methods.

Advanced Confocal Laser Scanning and Super-Resolution Microscopy System

1. Introduction and Purpose

Brookhaven National Laboratory (BNL) is planning to purchase an Advanced Confocal Laser Scanning and Super-Resolution Microscopy System, named herein super resolution microscope system (SRM), that is capable of high-speed, multi-color fluorescence imaging, including fluorescence lifetime (FLIM) and superresolution nanoscopy with Stimulated Depletion Emission (STED).

2. General Description

The SRM system will

Combine confocal, FLIM, and STED nanoscopy modalities in a unified platform.

Provide high sensitivity for live-cell imaging and long-term time-lapse studies.

Offer tunable excitation across a broad spectral range 450–780 nm and detection from 400nm up to 850 nm.

Enable lifetime-based functional imaging and spectral separation to distinguish overlapping fluorophores.

Integrate automation features, intuitive software, and full compatibility with standard imaging analysis workflows to provide hands free operation.

Provide both software based and hardware based super resolution modes of imaging.

3. Principal Configuration and Performance Requirements

3.1 Optical and Illumination System

The SRM will have a pulsed White Light Laser with high stability, variable repetition rate equipped with a pulse picker or similar and covering 450–780 nm and with the ability to simultaneous select various laser lines for optical excitation. The system will allow software-based control of laser intensity for each individual laser line, including when lines are used simultaneously.

Attachment B

The SRM will be provided with the option of secondary laser ports supporting UV (405

nm) and STED depletion lasers (598, 660, 775 nm).

The SRM will have high-speed beam routing for multicolor excitation switching within microseconds.

The SRM will have a set of objective lenses from low mag to high mag including lenses capable of STED.

3.2 Detection System

4 The SRM will have a spectral detection range of around 410–850 nm, with up to five simultaneous, tunable detection channels with a spectral resolution in the form of adjustable emission bandwidth in 1 nm increments.

5 The SRM will have acoustic optical beam splitters for emission selection with bandwidth in 1 nm increments.

6 The SRM will utilize photon detectors with high efficiency, around 50% @ 500 nm, which is typical for hybrid photon detectors; provide photon detectors which support analog, photon counting, and reflection modes and a large dynamic range for linear photon counting, up to 100 million counts/sec.

7 The SRM will provide dark noise minimization for photon detectors via active cooling and electronic stabilization.

3.3 Lifetime and Functional Imaging

The SRM lifetime imaging platform should be capable of: (1) functional mapping of cell physiology (pH, ion concentration, metabolic state); (2) rejection of reflection and autofluorescence for increased signal; (3) lifetime-based component separation; (4) lifetime-based FRET or protein-protein interaction analysis; (4) fluorophore separation via lifetime super resolution STED with background suppression.

The SRM temporal precision should be such that it can have the ability to record and resolve lifetimes up to tens of nanoseconds with sub…
