About this file

This is a Performance Work Statement (PWS) for a NASA-sponsored research contract titled "Effect of Radiation Quality and Galactic Cosmic Radiation Simulation on the Risk of Carcinogenesis in an Outbred Mouse Model (HS/Npt)," sponsored by NASA's Human Research Program (HRP), Space Radiation Element (SRE). The performance period runs from April 1, 2026 to March 31, 2030, with a Principal Investigator to be designated by the contractor.

The primary objectives are to determine the effects of radiation quality, dose, and dose rate on cancer initiation, promotion, and progression in HS/Npt outbred mice; establish relative biological effectiveness (RBE) and quality factor estimates for space-relevant radiation exposures; provide bio-banked tissue and comprehensive metadata to support future NASA investigations; and optionally assess cardiovascular and central nervous system outcomes and sex-specific responses within budget constraints. The contractor shall design and execute experiments utilizing 3,500 outbred HS/Npt mice (1,750 per year for two years, including both males and females) at the NASA Space Radiation Laboratory (NSRL), including gamma irradiation, full galactic cosmic ray simulation (acute and fractionated), and single-ion exposures (e.g., 56Fe) with dose coverage relevant to Mars design reference missions (300–450 mGy). Required deliverables include evaluation of carcinogenesis RBEs with data on overall survival, solid tumor-free survival, leukemia/lymphoma-free survival, and tumor spectrum analysis; biobanking of specified organs and tissues with metadata; a comprehensive digital data management plan; annual interim progress reports at 12-month intervals; and a final report at project completion. NASA will provide animal breeding, shipping to NSRL, and beam time at no cost; the contractor shall budget for animal housing, travel, experimental execution, and sample processing. Primary points of contact are Jason Weeks (Space Radiation Element Manager), Joshua Hlavenka (NASA Contracting Officer), and Kat Jaime (NASA Contracting Specialist) at Johnson Space Center.

Performance Work Statement (PWS)

Project Title:

Effect of Radiation Quality and Galactic Cosmic Radiation Simulation on the Risk of Carcinogenesis in an Outbred Mouse Model (HS/Npt)

Sponsoring Organization:

NASA Human Research Program (HRP), Space Radiation Element (SRE)

Principal Investigator (PI):

[Insert Name]

Period of Performance:

April 1, 2026 to March 31, 2030

1. Background Astronauts on long-duration space missions are exposed to galactic cosmic rays (GCR) and solar particle events (SPE), creating elevated risks of carcinogenesis, cardiovascular disease (CVD), and central nervous system (CNS) decrements. Traditional inbred animal models have limitations due to genetic uniformity, potentially leading to overestimation of relative biological effectiveness (RBE). Outbred mouse models (HS/Npt) provide a genetically diverse system that better mirrors human populations and may yield more accurate assessments of radiation-induced cancer and related health risks.

2. Objectives The objectives of this study are to:

1. Determine the effects of radiation quality, dose, and dose rate on cancer initiation, promotion, and progression in HS/Npt outbred mice.

2. Establish RBEs and quality factor estimates for space-relevant radiation exposures.

3. Provide bio-banked tissue and comprehensive metadata to support future NASA investigations.

4. (Optional) Assess cardiovascular and CNS outcomes, as well as sex-specific responses, within budget constraints.

3. Scope of Work The PI shall design and execute experiments utilizing outbred HS/Npt mice supplied by NASA. Work shall include:

- Radiation exposures at the NASA Space Radiation Laboratory (NSRL)(See Attachment C – NSCOR NSRL Information), including gamma irradiation, full GCR simulation (acute and fractionated), and single-ion exposures (e.g., 56Fe) where justified (See Attachment D – GCR Sim Table).

- Dose coverage relevant to Mars design reference missions (300–450 mGy).

- Survival, tumor incidence, and tumor spectrum analyses.

- Comprehensive tissue collection, preservation, and metadata management.

- Biobanking of required samples per NASA specifications.

- Preparation of interim and final reports.

- Mice availability in table below

Year Mice Numbers 1 1750 2 1750 Total 3500

Th majority of animals will be available in the first two years of the study due to the need to age the mice post irradiation. Mice would include both males and females.

4. Deliverables Required Deliverables:

- Evaluation of carcinogenesis RBEs for GCRsim and reference ions, including (provided in the annual reports below):

* Overall survival

* Solid tumor-free survival

* Leukemia/lymphoma-free survival

* Tumor spectrum/type analysis

- Biobanking of specified organs/tissues with metadata.

- Comprehensive digital data management plan covering animal, sample, and metadata tracking.

- Annual Reviews: Interim progress reports to NASA at 12-month intervals

- Final Report: Delivered at project completion (Month 48)

Optional Deliverables:

- Assessment of cardiovascular injury from space-like radiation exposure.

- Evaluation of CNS functional/physiological decrements.

Attachment A 80JSC026Q0003 Page 3 of 3

- Assessment of dose-rate effects.

- Analysis of sex-specific responses.

5. Required Tissue Collection and Preservation The PI shall collect and preserve biospecimens as outlined in NASA requirements (See Attachment B - Tissue Collection Description). Methods include snap freezing, paraffin embedding, or ethanol preservation, depending on tissue type. All required samples and metadata shall be transferred to NASA at the conclusion of the performance period.

6. Budget and Funding

- NASA will provide animal breeding, shipping to NSRL, and beam time at no cost to the PI.

- The PI shall budget for animal housing, travel, experimental execution, and sample processing.
