Statement of Work (SOW) Project: Validate Causative Mutations in Agriculturally Important Vertebrates
GENERAL INFORMATION
1.0 Scope of Work:
Provide a Cell Analyzer Brand Name (or Equal) to: BD Accuri C6 Flow Cytometer System.
2.0 Background:
The overall goal is to phenotype genome-edited cells to identify improved traits in pigs and poultry leading to increased production, improved animal welfare enhancing farm profitability and sustainability. Genome editing to produce changes in DNA thought to enhance production agriculture traits will be analyzed by culturing edited cells in vitro to determine if there is a positive effect on cell phenotype. The objective of this work is to develop poultry and livestock genotypes that will provide improved disease resistance traits to viral and/or bacterial pathogens. A new Research Virologist position at USDA-ARS was recently filled. The scientist will use gene editing and other technologies to systematically alter DNA sequences predicted to improve traits to cause beneficial changes in poultry and livestock production and test these changes in vitro and in vivo where warranted. The scientist will investigate and identify host variants associated with disease susceptibility or resistance. The outcome will be improved understanding of host dependencies of pathogens for both poultry and livestock. The scientist will contribute towards identifying and establishing poultry and porcine cell lines that can be used to phenotype agriculturally important traits in vitro while identifying the effects of mutations on these cell lines. Poultry and livestock genotypes that will provide improved disease resistance traits to viral and/or bacterial pathogens will be developed. The cell analyzer with live-cell imaging offers real-time cellular analysis and evaluates cell numbers, cell viability, apoptosis induction, morphology, cell health, proliferation, and cell-mediated toxicity.
CONTRACTOR REQUIREMENTS
3.0 Technical Requirements/Tasks:
• The cell analyzer should be Brand Name (or Equal) to BD Accuri C6 Flow Cytometer System
• The cell analyzer has a minimum of 2 standard lasers (a blue and red laser) offering simultaneous parameter detection for maximum coverage of the fluorochrome emission spectrum. The system should be equipped with two scatter detectors, and four fluorescence detectors with interference filters optimized for the detection of FITC, PE, PerCP-Cy™5.5, and APC
• Should have 200-μm ID quartz flow cell with minimum detectable particle size of 0.5 μm with a minimum sample volume of 50 μL
• Each detector array has to be equipped with appropriately matched optical filters for light collection
• A sheath focused core enables event acquisition rates of up to 10,000 events per second and a sample concentration over 5 x 106 cells per mL
• Should offer a fluorescence sensitivity of FITC <75; PE <50; fluorescence linearity 2 ±0.05% for chicken erythrocyte nuclei (CEN) and fluorescence precision <3% CV for CEN
• Should include tubing peristaltic pump, sheath additive, flow cell clean solution, test beads.
• Must include laptop with software compatible with device
• Must include installation
• Must provide 1-year extended warranty.
