About this opportunity

The National Human Genome Research Institute (NHGRI) seeks cooperative agreement applications to establish the GREGoRi Technology Integration Center (TechIC) supporting rare disease diagnosis through emerging molecular technologies, with applications due October 30, 2026 on a rolling basis for review cycles in March 2027, May 2027, and July 2027. One award of $3 million in total FY 2027 funding is anticipated, with a maximum project period of five years and a direct-cost budget cap of $2.25 million per project year.

Eligible applicants include domestic higher education institutions (public and private), nonprofits (501(c)(3) and non-501(c)(3)), for-profit organizations, state and local governments, tribal organizations and governments, school districts, special district governments, public or Indian housing authorities, and other domestic entities; foreign organizations and non-domestic components of U.S. organizations are not eligible, though foreign subawards are permitted. The PD/PI must commit at least three person-months annually (or 1.2 months if multiple PDs/PIs with combined effort of at least three person-months).

The TechIC will develop a structured, multi-omic dataset from individuals with rare genetic diseases and controls to enable benchmarking of molecular assays beyond DNA sequencing. Core deliverables include establishing the dataset with whole genome sequencing, DNA methylation, chromatin accessibility, transcriptome, and proteomic/metabolomic data; developing methods and best practices for applying these technologies in clinical genetics; and partnering with the GREGoRi Data Coordination and Outreach Center to share harmonized data via NHGRI's AnVIL platform. Applications involving human subjects research require IRB approval and informed consent for broad data sharing without additional restrictions. The center must participate actively in GREGoRi Consortium governance, including Steering Committee and working group meetings, and collaborate with related initiatives such as IGVF, HPRC, and ClinGen.

This opportunity operates under CFDA 93.172 (Human Genome Research) using the U01 cooperative agreement activity code.

Synopsis

The Genomics Research to Elucidate the Genetics of Rare Diseases:innovation (GREGoRi) initiative seeks to accelerate a paradigm shift in rare disease diagnosis by reimagining the tools, molecular technologies and analytical approaches used to identify the causal gene(s) and/or variant(s) underlying rare genetic disorders. The purpose of this Notice of Funding Opportunity is to establish the GREGoRi Technology Integration Center, which will enable the development of standards and best practices for applying new and emerging molecular methods in rare disease diagnosis. A major deliverable of the Center will be a multidimensional dataset that can be used for the development and benchmarking of novel tools and strategies that facilitate rare disease diagnosis.

Eligibility

Refer to Section III. Eligibility Information in the NOFO for additional information on eligibility.Foreign Organizations/International Collaborations:Non-domestic (non-U.S.) Entities (Foreign Organizations) are not eligible to apply.Non-domestic (non-U.S.) components of U.S. Organizations are not eligible to apply.Foreign components, as defined in the NIH Grants Policy Statement, are allowed.
