About this opportunity

The National Institutes of Health National Human Genome Research Institute (NHGRI) is soliciting applications for cooperative agreements to fund proof-of-concept studies exploring novel strategies for identifying causal genes or variants underlying rare genetic disorders using new and emerging molecular technologies, with an application due date of October 30, 2026.

Eligible applicant types include higher education institutions (public and private), nonprofits (501(c)(3) and non-501(c)(3)), small businesses, for-profit organizations, state and local governments, tribal governments and organizations, and foreign organizations and non-domestic components of U.S. organizations. All applicants must register with SAM.gov, obtain a UEI, and establish eRA Commons accounts before submitting through Grants.gov.

The NHGRI will award 8–10 cooperative agreements totaling $7.5 million in FY 2027, with a ceiling of $500,000 in direct costs per year for a maximum three-year project period. No cost share is required.

The initiative seeks to move beyond DNA sequencing-based initial diagnostic approaches. Two categories of innovation are eligible: Technology Innovation Projects conducting proof-of-concept studies using emerging experimental technologies (multi-omics, RNA sequencing, metabolomic analyses), and Computational Innovation Projects developing novel analytical methods and user-friendly tools for variant identification. Work will be performed at applicant sites with administrative and collaborative oversight through the GREGoRi Research Consortium. Awardees must operate as consortium members, collaborate with consortium and related initiatives (including IGVF and HPRC), and share all data, metadata, protocols, and research products with the GREGoRi Data Coordination Center and through AnVIL with broad research access.

The application opens September 30, 2026. Scientific merit review is scheduled for October 2026, advisory council review for March 2027, and the earliest start date is July 2027. Awards are made under CFDA 93.172 (Human Genome Research).

Synopsis

The Genomics Research to Elucidate the Genetics of Rare Diseases:innovation (GREGoRi) initiative seeks to accelerate a paradigm shift in rare disease diagnosis by reimagining the tools, molecular technologies and analytical approaches used to identify the causal gene(s) and/or variant(s) underlying rare genetic disorders. This Notice of Funding Opportunity is intended to stimulate the development and testing of highly innovative experimental or computational approaches for rare disease diagnosis, that have the potential to make transformative improvements to the current state of the art.

Eligibility

Refer to Section III. Eligibility Information in the NOFO for additional information on eligibility.Foreign Organizations/International Collaborations:Non-domestic (non-U.S.) Entities (Foreign Organizations) are eligible to apply.Non-domestic (non-U.S.) components of U.S. Organizations are eligible to apply.Foreign components, as defined in the NIH Grants Policy Statement, are allowed.
