About this opportunity

The Department of Health and Human Services National Institutes of Health Office of Logistics and Acquisition Operations seeks procurement of a Tokai Hit STXG-GSI2X-SET stage top incubator for use with an existing Leica Stellaris 5 STED super-resolution microscope, with proposals due July 25, 2026. The procurement is a Total Small Business set-aside.

Place of performance is Bethesda, Maryland. The incubator must provide precise dynamic control of temperature, humidity, CO2, and oxygen for live cell culture imaging. Technical specifications require dual feedback modes (chamber temperature or specimen temperature) with temperature control from 25 to 50 degrees Celsius and continuous room temperature monitoring. The built-in digital gas mixer controller must accept 100% CO2 from lab supply and regulate CO2 concentration between 5.0 and 20% at the sample with real-time sample feedback. Active humidity control via internal humidifier is required. The electrically heated stage top incubator chamber must feature magnetic specimen holder interchanges compatible with 35 mm coverglass dishes, 50/60 mm coverglass dishes, chamber slides, coverglass chambers, and multi-well chamber frames. A flexible heated collar must accommodate variable objective lens diameters. The unit must measure 112 x 87 mm, be compatible with the Leica Super Z galvo stage, weigh less than 140 grams, and operate on 110–240V power. The system must include an external sensor for autoclavable repeatability and sample feedback regulation, and feature Tokai Hit original data logging software with temperature and CO2 program control compatible with LAS-X software. NAICS code is 334516, Analytical Laboratory Instrument Manufacturing; PSC is 6640, Laboratory Equipment and Supplies.

Requirements:

Provides accurate dynamic control of temperature, humidity, CO2 and oxygen to maintain required cell culture conditions for live cell confocal and STED super-resolution imaging. The stage top incubator is required in order to perform live cell imaging with the unique resolution capabilities of our Leica Stellaris 5 STED microscope. It currently does not have a stage top incubator, and therefore, is unable to perform this type of imaging requested by NCI researchers.

Stage top incubator controller must have two feedback modes, chamber temperature or specimen temperature, while it also continuously monitors the room temperature to maintain specimen temperature within set point anywhere between 25 – 50 degrees Celsius. The built-in digital gas mixer controller accepts 100% CO2 from lab supply to regulate CO2 concentration at the sample, with a CO2 range of 5.0 to 20% with real-time sample feedback. In addition, humidity is actively controlled with an internal humidifier to sense sample chamber humidity. The electrically heated stage top incubator chamber must have magnets for easy interchange of specimen holders, including inserts for 35 mm coverglass dish, 50/60mm coverglass dish, chamber slide, coverglass chamber, and open frame insert for imaging multi-well chambers. A flexible heated collar that accommodates a range of objective lens diameters must also be included.

The components of the stage top incubator must include:

  • Tokai Hit STXG-GSI2X-SET Stage Top Incubator for Super Z galvo stage (inverted microscope stands)
  • Pure (100%CO2) gas use (110- 240V) for individual temperature settings to create optimum temperature and humidity conditions without difficulty for long term live cell imaging.
  • Setting temperature range from 25 to 50°C.
  • Dimensions of 112 x 87 mm compatible with Leica Super Z galvo stage on existing Leica Stellaris 5 STED microscope.
  • the lightest chamber (less than 140g) for Super Z Glavo stage
  • temperature controller with built-in digital gas mixer (CO2: 5.0 - 20.0%)
  • Objective heater
  • Magnetic Dish holder and fixing lid for 35mm, 50/60mm dish, chamber slide, slide glass and chamber slide use.
  • External Sensor, enable to autoclave repeatability for Sample Feedback regulation.
  • Tokai Hit original data logging, temp/CO2 program control, software compatible to use with LAS-X software
