About this opportunity

The Department of Health and Human Services' National Institutes of Health is conducting market research through a Sources Sought notice to identify qualified sources capable of providing a Covalent Screening Library of Small Molecules consisting of 1,000 compounds (500 from Sulfamoyl Fluorides set and 500 from Cyanamides set). The procurement falls under NAICS code 32599 with a size standard of 650 employees and aims to expand the National Center for Advancing Translational Sciences (NCATS) Experimental Therapeutics Branch's chemical compound collection to target previously undruggable proteins and enhance quantitative high-throughput screening campaigns. Key requirements include validated biological and pharmacological activities, compatibility with high-throughput and high-content screening, detailed biological and chemical information with LC/MS and/or NMR reports, and guaranteed purity exceeding 90 percent. Companies must submit capability statements that specifically address each project requirement, include total employee count, professional qualifications, GSA Schedule or GWAC contracts if applicable, and demonstrate technical competence in manufacturing these specialized chemical compounds. The response deadline is August 11, 2025, at 10:00 AM EDT, with submissions required electronically to Contract Specialist Diedra Prophet and Contract Officer Christine Frate.

This Sources Sought notice does not include any set-aside designations and encourages participation from all qualified businesses, including small businesses. No incumbent contractors or potential award values are mentioned in the documentation, as this is strictly a market research effort to determine supplier availability and capability rather than a formal solicitation. The procurement specifies a quantity of exactly 1,000 small molecules divided equally between two distinct chemical sets, with delivery required within six weeks after purchase order receipt. Companies responding must identify their business type and size status under the applicable NAICS code, with small businesses required to address Non-Manufacturer Rule compliance regarding the actual manufacturer's size status. The government has explicitly stated that this notice does not obligate any contract award and that responses will not be considered as formal proposals or quotes, with no costs reimbursed to responding companies.

Title: Covalent Screening Library of Small Molecules (500 from Sulfamoyl Fluorides set and 500 from Cyanamides set)

Classification Code: 6810 - CHEMICALS

NAICS Code:  32599-ALL OTHER MISCELLANEOUS CHEMICAL PRODUCT AND PREPARATION MANUFACTURING

Size Standard: 650 employees

Description:

This is a Sources Sought notice. This is NOT a solicitation for proposals, proposal abstracts, or quotations. The purpose of this notice is to obtain information regarding the availability and capability of all qualified sources to perform a potential requirement.

This notice is issued to help determine the availability of qualified companies technically capable of meeting the Government requirement, to determine the method of acquisition, and availability of domestic sources manufactured in the United States in sufficient and reasonably available commercial quantities and of a satisfactory quality.  It is not to be construed as a commitment by the Government to issue a solicitation or ultimately award a contract.  Responses will not be considered as proposals or quotes.  No award will be made as a result of this notice.  The Government will NOT be responsible for any costs incurred by the respondents to this notice.  This notice is strictly for research and information purposes only. Small businesses are encouraged to respond.

For equipment/supply requirements, responses must include the place of manufacturing (i.e. address if supply/equipment is a domestic end product and include country of manufacturing). For equipment/supply requirements, small businesses must also address the size status of the manufacturer under the applicable NAICS code (i.e. address Non-Manufacturer Rule).

Background: The current DPI compound collection notably lacks a collection of custom covalent libraries. Covalent inhibitors offer several advantages, including the ability to target proteins considered undruggable in live cells, the development of novel affinity-based probes, enhanced potency, and prolonged duration of action. This approach is particularly appealing in both medicinal chemistry and chemical biology research. Acquiring these compounds from Enamine is imperative to development of a focused, custom covalent library that spans a broader and more diverse chemical space. Screening compounds with reactive warheads has emerged as a promising strategy to both discover and validate new targets, and to identify hits for targets that are otherwise considered undruggable. Acquiring such a library will provide complete access to resources for DPI groups, thereby advancing their programs and enhancing or complementing their quantitative high-throughput screening (qHTS) campaigns.

Purpose and Objectives: NCATS ETB is looking to expand its covalent screening library of small molecules. Adding diversity to chemical libraries is imperative to our operation, as we screen a variety of targets against these in-house collections.
