Statement of Work (SOW)
Project: Validate Causative Mutations in Agriculturally Important Vertebrates
GENERAL INFORMATION
1.0 Scope of Work:
Provide a CO2 incubator.
2.0 Background:
The overall goal is to phenotype genome-edited cells to identify improved traits in pigs and poultry leading to increased production, improved animal welfare enhancing farm profitability and sustainability. Genome editing to produce changes in DNA thought to enhance production agriculture traits will be analyzed by culturing edited cells in vitro to determine if there is a positive effect on cell phenotype. The objective of this work is to develop poultry and livestock genotypes that provide improved disease resistance to viral/bacterial pathogens and/or other performance related traits. A new Research Geneticist position at USDA-ARS was recently filled. The scientist will use gene editing and other technologies to systematically alter DNA sequences predicted to improve traits to cause beneficial changes in poultry and livestock production and test these changes in vitro and in vivo where warranted. The scientist will contribute towards identifying and establishing poultry and porcine cell lines that can be used to phenotype agriculturally important traits in vitro and evaluate the effects of natural genetic variation or novel mutation on corresponding cellular phenotypes. The CO2 incubator provides stable growth environments for different types of cells by controlling the CO2 and oxygen levels thereby maintaining correct pH in the growth cultures.
CONTRACTOR REQUIREMENTS
3.0 Technical Requirements/Tasks:
• The CO2 incubator shall have165L (5.8 cu. ft.) capacity copper tri-gas single chamber with adjustable, perforated shelving and have HEPA filtration Steri-
Run sterilization cycle.
• The CO2 incubator must provide a temperature range of +3° to 55°C, 5-90% O2 control, and 1-20% CO2 concentration range with a bulb free IR CO2 sensor (IR
Sensor Technology).
• The CO2 incubator must have a directly heated, covered integrated water reservoir providing stable relative humidity >93% at 37°C and offer a condensation free inner chamber with fan distributing clean, humidified air throughout the chamber.
• The CO2 incubator must offer bright, door-mounted display for easy accessibility and viewing.
4.0 Deliverables / Schedule:
