Statement of Work Project: Mucosal colonization of Mannheimia haemolytica and isogenic mutants in cattle.

GENERAL INFORMATION

1.0 Scope of Work:

Provide 12 Colostrum Deprived Holstein or Holstein/cross Calves.

2.0 Background:

Bacterial bovine respiratory disease continues to be the most costly disease-related issue facing the US cattle industry. Control of the disease is currently accomplished primarily through the extensive use of antibiotics. Vaccine control measures are widely available but suffer from relative inefficacy, in part due to problems with timely delivery. Bacterial capsule is prominent among these features, inhibiting phagocytic activity and complement-mediated killing. Sialic acid decoration of LPS appears to be an immunologic stealth mechanism for P. multocida (and other organisms) whereby the bacterium can escape immune detection.

Our objective is to evaluate the nasopharyngeal colonization of a wildtype Mannheimia haemolytica and seven isogenic mutants (capsular, sialic acid, filamentous hemagglutinin, leukotoxin, and three adhesin mutants) in cattle.

Determine what role, if any, inactivation of specific regions of Mannheimia has on the observed high-level colonization of wild type Mannheimia; the effect on colonization during a concurrent virus infection.

CONTRACTOR REQUIREMENTS

3.0 Technical Requirements/Tasks:

We require the following of the calves:

• Must be BVD-negative for antibody and antigen

• Must be Mycoplasma bovis negative

• Colostrum Deprived

• No milk replacer for the first 48 hours

• Housed individually

• Must be dewormed

• Must be free of ring-worm

• Must be sound of foot and body

• Must be 7-8 weeks old

• Animals must be transported directly to NADC with no other animals in the trailer.

• Must have 12 calves available with comparable size across the group of the same breed (mixed dairy/beef breed)

• Certificate of Veterinary Inspection required on all animals.

• Calves shall demonstrate no signs of clinical illness.

• Calves shall have sound legs and good eyes.

• Calves must be sourced from a farm within 100 miles of Ames to limit transport time of the animals on the trailer.

• Blood work will be required from the time of award until the arrival of animals as needed to check for BRSV and Mycoplasma titer level.

• Communication from the time of award to the arrival of animals must be had between Animal Resources Unit in Ames and the possible vendor. This communication may include group or individual animal health status, blood work results, housing and feeding information that could be subject to change within this time frame.

• Animal Resources Unit may exclude calves prior to arrival that do not meet the stated requirements.

• To obtain 12 animals that meet all specifications; the vendor may need to obtain a group larger than 12 to allow for alternate animals to replace any that become sick or do not meet the health requirements.

4.0 Technical Point of Contact:

5.0 Delivery Location:

To USDA, 1920 Dayton Avenue, Ames, IA 50010 within 8 weeks of award.
