Statement of Work (SOW) Project: Purchase of microtome for tissue sectioning

GENERAL INFORMATION

1.0 Scope of Work: Dairy cattle have varying feed efficiency and production values based on their microbiome, but we are not sure what shapes their microbiome, genetics, environment, or a mix of both. Environment can be difficult to manipulate but genetics are permanent and cumulative and often result in a visible phenotype. The goal of this study is to identify the height, width, and space between various papilla found in each compartment of the rumen. The first objective has been met and tissue has been collected and is in formalin. The second objective is to have tissue imbedded in paraffin, and placed in blocks, followed by cutting samples and staining the samples. Having the capability to cut our own blocks in the thickness we want will give us more opportunities to test various stains on the samples.

2.0 Background: We require a microtome that can cut thin (greater than or equal to 3μm) sections of paraffin imbedded tissues. Currently we do not own a microtome at DFRC and I foresee this being a reoccurring process for us as we untangle the web of genomic vs environmental effects on the rumen microbiome. We have an excellent microscope that will be put to good use in measuring the papilla and the spaces between papilla once tissues are sectioned. In addition to the currently planned studies this would give us an opportunity to section other items such as forages to look for cellular infiltrates and mammalian tissues other than rumen tissues.

CONTRACTOR REQUIREMENTS

3.0 Technical Requirements/Tasks:

1. The microtome must be able to cut paraffin embedded samples at a depth of greater than or equal to 3 microns

2. The microtome must have a mechanism to lock the handwheel while the object and knife are being mounted and a finger guard for safety

3. The microtome must have a color-coded tangible zero point setting

4. Themicrotome must be able to be used with large universal cassette clamps allowing different sized samples to be processed

5. The microtome must be easy to clean and have an ergonomic design

6. The microtome must have a backlash- and maintenance-free horizontal and vertical cross-roller bearing system

7. The microtome must have a quick change system for object clamps

8. The microtome must have an electronic touch screen that has a memory function and section counter

9. The microtome must be able to cut sections between 0.5-100 μm

10. The microtome must have a coarse feed speed of 1.0mm/sec

11. The microtome must have multiple setting ranges
