About this file

This statement of work outlines requirements for adding solid phase microextraction (SPME) capabilities to an existing gas chromatography-mass spectrometer with autosampler. The USDA Agricultural Research Service Midwest Area seeks to integrate new metabolomics data with other omics datasets to develop networks associated with barley malting quality. The selected vendor will provide a smart SPME fiber kit including SPME tool, fiber assortment kit, and performance evaluation mix, as well as SPME liners and installation/training services. The equipment must be delivered by the end of fiscal year 2023 to the USDA ARS Cereal Crops Research Unit in Madison, Wisconsin. The agency will provide a one-year manufacturer warranty for the delivered solution.

Statement of Work (SOW) Adding SPME Capabilities to an existing GCMS-TQ8040 with an AOC-

6000 Autosampler

OR EQUIVALENT

Dr. Sarah Whitcomb

USDA ARS Cereal Crops Research Unit 502 Walnut St.

Madison, WI 53726

GENERAL INFORMATION

1.0 Scope of Work: In this project we will conduct a detailed metabolome analysis of raw barley kernels, barley malting stages, and barley malt extract. We will integrate these new metabolomics data with other omics datasets (transcriptome, proteome, miRNome) previously generated by the research unit to develop cogent molecular networks associated with barley malting. These networks will aid the identification of novel enzymes and regulatory factors (hub genes) associated with malting quality. Further, the metabolomic data generated will be used to identify new parameters and traits that can influence and/or predict malt quality and character.

This addon to our current autosampler will allow us to analyze the headspace of our samples more affectively for select compounds and allow us to save preparation time for these samples.

2.0 Background:

The Whitcomb laboratory currently does not have a SPME fiber capability on our GCMS autosampler. We will utilize this new method on an almost daily basis for the foreseeable future. This addon to the autosampler will give us a new method to analyze the headspace of our samples in a way that we currently don’t have.

Below is a list of technical specifications that are best suited for our experimental needs:

• Smart SPME Fiber Kit o SPME tool (1 piece).

o Smart SPME Fiber Assortment Kit(1 piece).

o SPME Performance Evaluation Mix(1 piece).

• SPME Liner o SPME liner 0.75mm x 5.0mm x 95mm for Shimadzu GC’s, 5pk

• Tier 1 Installation and Customer Familiarization

4.0 Date Needed
