Statement of Work Project: Assessment of the immunogenicity and pathogenicity of M. avium subspecies paratuberculosis mutants in calves
GENERAL INFORMATION
1.0 Scope of Work: The overall objective of this study is to determine the immunogenicity and pathogenicity of two live-attenuated mutants of
Mycobacterium avium subspecies paratuberculosis (Map). These two mutants are promising vaccine candidates for Johne's disease. Calves will be used to test these live vaccines. The vaccines were initially constructed based on the immunogenic properties of the gene knockouts (see Bannantine et al., 2011). Therefore, we hypothesize that the vaccines will both be immunogenic and may protect against
Johne's disease.
2.0 Background: Map causes an economically significant disease to the dairy industry in the US and worldwide. Currently, there is no effective vaccine for Johne's disease and vaccination is considered the best control strategy for this disease. If the live-attenuated mutants tested in this study activate protective immune responses, it could go a long way to reducing the negative impact of Johne's disease on national/international trade as well as help farmers better manage their herds.
