STATEMENT OF NEED/TECHNICAL SPECIFICATIONS

DNA Magnetic Bead Extraction Instrument with supplies

1333LF21QNFFK0059

1.0 BACKGROUND

The NWFSC Marine Forensics lab provides casework analysis for NOAA Office of Law Enforcement (OLE). OLE relies on the Marine Forensic Laboratory to provide DNA and morphological analyses, written reports, and expert testimony in support of criminal and civil investigations. OLE is charged with enforcing federal laws on endangered species take, seafood mislabeling, and Illegal, Unreported, and Unregulated (IUU) Fishing. This request is to purchase two automated DNA extraction instruments and components in order to maintain the high quality and timely work product for NOAA Fisheries.

1.1 OBJECTIVE

The purpose of this request is to purchase two automated DNA magnetic bead extraction instruments with supplies for the NWFSC Marine Forensic Laboratory in Seattle, WA and in Charleston, SC. We require these DNA extraction instruments to maintain and improve our capacity for processing NOAA Office of Law Enforcement (OLE) Forensic DNA casework. The current DNA extraction instruments in the forensic labs are out-of-date and no longer supported by the manufacture. We need to replace our out-of-date DNA extraction instruments to prevent a disruption in casework analysis for NOAA OLE. In addition, we require the same DNA instrument and components across our two labs to increase our sample throughput and to standardize forensic DNA extraction protocols.

2.0 TECHNICAL SPECIFICATIONS The

We require a genomic DNA extraction instrument with the following specifications to meet our needs:

• Automated and hands-free genomic DNA purification robotic instrument using magnetic bead DNA capture technology

• Bench top instrument that can fit inside our existing laboratory bench space, no larger than 40” x 28” x 36”

• Allows for external, enzymatic sample preprocessing and variable input volumes

• Allows for variable elution volumes

• Can process up to 96 sample a day or more

• Can process variable number of samples with a minimum of 2 and maximum of 24 in one run.

• Can process variable number of samples without excessive plastic waste from unused wells from a plate

• Can batch process variable number of samples in the same run using the standard spacing 8 well column/plate format

• Can elute DNA into standard 8 column wells (strip tubes) or 96 well plates to be compatible with our existing downstream processes and storage. For example, the instrument does not elute ONLY into a single 1.5 milliliter tubes.

• Can handle multiple types of biological material for DNA extraction including one for muscle tissue protocol.

• No cross contamination detected between DNA samples

• UV decontamination and a closed housing

• Software with audit trail documentation

• Includes shipping and on-site instrument installation plus initial training

• Includes a one-year warranty

The following components, reagents, accessories, and supplies are required to operate the automated DNA magnetic bead extraction instrument from Roche Diagnostics or equal.

• 2 x 07652275001 FrameStrip with flat caps-High profile

• 2 x 07345577001 MagNA Pure 24 or equal Processing Cartridge

• 2 x 07345585001 MagNA Pure 24 or equal Tip Park/Piercing Tool

• 8 x 07658036001 MagNA Pure 24 or equal Total NA Isolation Kit

• 2 x 06241620001 MAGNA PURE 96 FILTER TIPS or equal (1000UL)

• 2 x 07857551001 MagNA Pure Tube 2.0mL or equal
