Purchase of confocal laser scanning inverted microscope
GENERAL INFORMATION
1.0 Scope of Work: Vendor is to deliver a new confocal laser scanning inverted microscope as identified in this solicitation.
Features, Components, Accessories, Modules, Software and/or Services to be included:
1) Inverted microscope platform with fully motorized scanning stage, remote touchscreen display, and autofocus
2) Objectives: Plan-Apochromat (PApo) 10x/0.45 numerical aperture, PApo 20x/0.8, PApo 40x/1.3 Oil DIC, PApo 63x/1.4 Oil DIC, and PApo 40x/1.2 DIC for water, silicon oil, or glycerine immersion.
3) Additional transmitted light detector and LED fluorescence illuminator
4) Laser modules: 405, 488, 561, and 640 nm
5) Gallium arsenide phosphide (GaAsP) photomultiplier tube (PMT) detectors
6) Software that supports Z-stack, time lapse, photobleaching/photoactivation, spectral unmixing, automated image analysis, measurements, colocalization, autofocus, 3D rendering, tile stitching, and deconvolution
7) PC workstation and monitor included
8) Anti-vibration table and air compressor
9) Installation and training.
2.0 Background: A confocal laser scanning microscope is required to perform imaging of live and fixed samples (e.g. fungi, yeast, plant, bacteria, host-microbe interaction and single molecules/nanoparticles), and specialized imaging techniques for localization samples, complicated biochemical labeling/ fluorescent dye selections that demand peak sensitivity and resolution from a point-scanning approach.
CONTRACTOR REQUIREMENTS
3.0 Technical Requirements/Tasks:
Technical Specifications:
1) Inverted microscope platform with fully motorized scanning stage, remote touchscreen display, and autofocus (smallest increment 10 nanometer)
2) Microscope must have the ability to perform Differential Interface Contrast (DIC), phase contrast microscopy, and switch between LSM and cameras
3) Microscope must support the addition of stagetop incubator for regulation of temperature, carbon dioxide, and humidity
4) Scanning stage 130x100 STEP with stepper motor having travel range of at least 130 mm x 100mm, 0.1 micrometer resolution, reproducibility of +/- 1 micrometer, and accuracy of +/- 5 micrometers
5) Objectives: Plan-Apochromat (PApo) 10x/0.45 numerical aperture, PApo 20x/0.8, PApo 40x/1.3 Oil DIC, PApo 63x/1.4 Oil DIC, and PApo 40x/1.2 DIC for water, silicon oil, or glycerine immersion.
6) Additional transmitted light detector and LED fluorescence illuminator (385 nm)
7) Three gallium Arsenide Phosphide-PMT (GaAsP-PMT) detectors supporting three channels, up to 8 frames/s @ 512 x 512 pixel and maximum frame resolution of 6144x6144 pixel.
8) Microscope must have the ability to be upgradable to a higher sensitivity detector.
9) Lasers: Diode 405nm (5mW), 488nm (10mW), 561nm (10mW), 640nm (5mW)
10) Filters for detection of reflected light fluorescent illumination, apochromatic corrected beam path with transmission, DAPI (Hoechst, BFP, Indo-1), GFP (EGFP, Alexa 488, CFP, FITC), Alexa 546 (Cy3, DsRed, dTomato, Rhodamine)
11) Confocal Imaging Software (Windows 10) supporting Z‐stack, time lapse, photobleaching/photoactivation, spectral unmixing, automated image analysis, measurements, colocalization, autofocus, 3D rendering, tile stitching, deconvolution, and all common file formats (jpg, tiff, czi, lsm, bmp, avi)
12) Software capabilities to use the motorized XY stage for capturing and stitching large fields of view, or automating the collection of marked XY positions; and creating a time series with different intervals over time, or multi-position Z-stacks that all different in the number of Z planes
13) Workstation PC capable of controlling instrument and confocal imaging software. Specification should be equivalent or better than the following:
High-performance processor (4 cores), Memory (16 GB DDR4-2666), 2 TB hard drive, Graphics Card (8 GB GDDR5 memory, PCR Express 3.0 Interface x 16, display support for 4x 32” display), 32” monitor
14) Anti-Vibration Table with low noise air compressor, 3.5 Liter Capacity, 110v
15) Installation and setup of instrument, workstation, and software.
16) One day of online and/or onsite training within 1 month of installation unless both parties agree on a different time.
17) 1 yr comprehensive warranty service coverage
4.0 Government Furnished:
Government will be present during delivery and installation of equipment. If needed, the government will supply a standard loading dock to accept delivery of the unit. The government will supply a location with suitable instrument requirements (space, electricity) as needed to operate the instrument. Equipment will be delivered to 1815 North University Street, Peoria, Illinois. Given CoVid restrictions, additional requirements will be in place for entrance to the lab. These will be communicated with the equipment supplier. Attempts will be made to ensure that safety is met for all parties and the equipment delivered in a timely manner.Buffers, water, bleach, and 70% ethanol for instrument cleaning will also be made available during servicing.
