About this file

This document is a requirement specification for a real-time qPCR thermocycler system and accessories for the USDA Agricultural Research Service (ARS) Pacific West Area (PWA) located in Albany, California. The specification details precise technical requirements for a research-grade PCR system, including a thermocycler that can generate a thermal gradient across a 96-well reaction block, support multiple fluorescent technologies, and capture up to five fluorophores in a single reaction well. The required system must have compatible software capable of data acquisition, viewing, and analysis for various PCR applications such as quantification, melt curve analysis, and gene expression analysis.

In addition to the primary thermocycler system, the specification calls for supplementary hardware for upstream sample processing, specifically a mini centrifuge for 0.5ml microcentrifuge tubes and a benchtop shaker incubator for sample agitation and incubation. The package should also include approximately 25mls of compatible qPCR mastermix containing fluorophore and enzyme. The document emphasizes that these requirements are essential for conducting mission-critical research involving quantitative PCR and upstream sample processing, indicating a high-precision scientific procurement for the USDA-ARS research facility.

Requirement specifications (Real time qPCR thermocycler system and accessories)

The following requirements are essential to conduct mission critical research involving quantitative PCR and upstream sample processing

Deliver to:

USDA - ARS - PWA

800 Buchanan Street Albany, CA 94710

• Thermocycler system must generate a thermal gradient across the 96-well reaction block to support rapid optimization of real-time PCR assays.

• Must be compatible with many fluorescent technologies for real-time PCR amplification.

• The System must capture up to five fluorophores in a single reaction well using “All Channels” scan mode, or, for faster scanning time, one fluorophore when using the SYBRR/FAM or FRET Scan Modes.

• Compatible software must supports data acquisition, viewing, and analysis of data generated by the system and supports real-time PCR applications such as PCR quantification, melt curve analysis, end point analysis, allelic discrimination, and gene expression analysis.

• Include approximately 25mls of compatible qPCR mastermix (fluorophore + enzyme)

• Additional hardware for upstream tissue processing and nucleic acid analyses are required: mini centrifuge for 0.5ml microcentrifuge tubes, benchtop shaker incubator for sample agitation and incubation.
