About this file
This Statement of Work (SOW) details a contract for analytical services for the U.S. Geological Survey (USGS) involving the analysis of fish liver samples from the Snake River. The objective is to analyze up to 80 fish liver tissue samples to determine concentrations of total microcystins using the 3-methoxy-2-methyl-4-phenylbutyric acid (MMPB) method, which can detect both free and bound microcystin forms. The contractor will provide analytical results in an electronic Excel spreadsheet or PDF, including sample identification numbers, microcystin concentrations, and measurement units, with delivery and invoice submission required by 03/31/2026.
The work stems from USGS long-term monitoring projects on juvenile fall Chinook salmon in Lower Granite Reservoir, where harmful algal blooms have occurred. The analysis will focus on detecting microcystins in liver tissue, as prior research indicates planktivorous fish accumulate higher toxin levels. The USGS will prepare and ship the liver tissue samples to the contractor's designated laboratory, with the contractor responsible for performing the specialized MMPB analytical method and reporting results. Acceptance criteria stipulate that all submitted samples must have corresponding analytical results provided in the required electronic format.

PR0044031882 Analysis Fish Liver Samples
STATEMENT OF WORK
1. BACKGROUND
The U.S. Geological Survey (USGS) conducts long-term monitoring projects on the Snake River to estimate abundance, growth, and survival of juvenile fall Chinook salmon migrating through Lower Granite Reservoir. A portion of these juvenile salmon remains in the reservoir for extended periods, during which they feed primarily on zooplankton and invertebrates.
In recent years, harmful algal blooms (HABs) have occurred during summer months in Lower Granite Reservoir. Prior research indicates that planktivorous fish accumulate higher levels of microcystins (MCs) than carnivorous fish and that liver tissue is a primary target organ for toxin accumulation.
This requirement represents a continuation of previously awarded analytical work supporting ongoing USGS monitoring efforts.
The 3-methoxy-2-methyl-4-phenylbutyric acid (MMPB) method is required to detect total microcystins (free and bound forms), as alternative screening methods may under-detect bound microcystins in liver tissue.
2. OBJECTIVE
Analyze fish liver tissue samples submitted by USGS to determine concentrations of total microcystins using the MMPB analytical method and to provide analytical results to USGS in an electronic format.
3. SCOPE OF WORK
The contractor shall analyze fish liver tissue samples provided by USGS in accordance with this Statement of Work.
3.1 Sample Quantity
Up to 80 fish liver samples for analysis.
3.2 Sample Preparation and Submission (Government-Furnished Items)
USGS will:
• Prepare liver tissue samples in accordance with contractor-provided procedures
• Individually package and label each sample with a sample identification number and sample type (liver tissue)
• Ship samples overnight on dry ice to the contractor’s designated laboratory
