About this file

This is a Statement of Work (SOW) for a procurement of consumables and reagents required to process 650 plasma exosomal RNA samples for next-generation sequencing analysis. The National Human Genome Research Institute (NHGRI), operating under the Department of Health and Human Services National Institutes of Health, seeks to characterize gene expression profiles of plasma exosomes to investigate the pathophysiology of cardiometabolic diseases across diverse populations. The project leverages molecular biology techniques to analyze plasma as a biological indicator of health and disease status, with the goal of identifying novel biomarkers and elucidating disease mechanisms. All consumables and reagents will be supplied by New England Biolabs (NEB) and are specifically selected for compatibility with the Illumina sequencing platform, which is the designated system available through NHGRI's supporting core laboratory.

The SOW emphasizes that the required products are essential for generating all necessary sequencing products and will support ongoing research efforts at the Center for Research on Genomics and Global Health. The procurement focuses on laboratory consumables and reagent supplies rather than services, with the technical specifications and quantities tailored to accommodate the processing of 650 plasma samples. This acquisition represents a targeted, specialized procurement for a defined research project with established analytical requirements and predetermined sequencing platform specifications.

Statement of Work

Transcriptomic characterization of plasma exosomes (NEB)

Background:

The Center for Research on Genomics and Global Health (CRGGH) explores how genetic and environmental factors contribute to the onset of complex diseases like Type 2 diabetes, obesity, and hypertension. Over the past two decades, CRGGH has gathered phenotypic data from more than 6,000 individuals both in the United States and internationally.

To better understand the underlying causes of metabolic disorders, CRGGH researchers utilize a variety of analytical methods. Over the years, they have systematically collected diverse types of -omics data, including genomics, transcriptomics, proteomics and microbiomes—from all or some participants, aiming to conduct integrated analyses that unravel underpinning mechanisms of these diseases.

Purpose and Objectives:

The primary purpose of this acquisition is to obtain consumables and reagents essential for processing 650 plasma exosomal RNA samples for next generation sequencing. The Center for Research on Genomics and Global employs various molecular biology techniques to investigate the pathophysiology of cardiometabolic diseases and associated traits across diverse populations. This project aims to characterize the gene expression profile of plasma exosomes, which are implicated in several diseases. Plasma, a biological fluid collected via minimally invasive methods, serves as an indicator of health and disease status. Molecular analysis of plasma may further elucidate disease mechanisms and facilitate the discovery of novel biomarkers.

Consumables and reagents supplied by New England Biolabs (NEB) are utilized in generating all necessary products for sequencing on the Illumina platform, the system currently available to NHGRI investigators through the core laboratory supporting this ongoing project.

-NEBNext® Multiplex Oligos for Illumina provide adaptors and primers to enable high yield multiplex Illumina library production. The unique hairpin loop structure of the NEBNext Adaptor minimizes adaptor-dimer formation, and NEBNext index PCR primers enable index incorporation during library amplification. It is characterized by high efficiency adaptor ligation, minimized adaptor-dimer formation and includes index primers for library multiplexing.
-The ribosomal RNA depletion kit is required for removing the ribosomal RNAs from the samples, thus eliminating the most abundant transcripts with minimal biological interest and background noise downstream of sequencing. This kit is compatible with a broad range of input amounts: 10 ng - 1µg and suitable for low-quality such as those extracted from specimen-like urine.
-NEBNext® Ultra™ II Directional RNA Library Prep with Sample Purification Beads kit is required for library preparation, an important step in next generation sequencing. This strand-specific RNA library prep kit has been reformulated at each step, resulting in several fold higher yields of high-quality libraries and enabling use of lower input amounts and fewer PCR cycles.

Period of Performance:

This is a one-time-use product, and the completion of the assays is expected to be completed within 1 year.
