About this file
This document is a Statement of Work (SOW) for a federal contract opportunity. The Agricultural Research Service (ARS) of the United States Department of Agriculture (USDA) is requesting Enzymatic Methyl-Seq (EM-seq) services, including DNA extraction, library preparation, and sequencing, for 106 tissue samples collected from cattle. The period of performance is from September 12, 2024 to December 31, 2024.
The solicitation is a small business set-aside under NAICS code 541380, with a size standard of $19M. Offerors must demonstrate technical capability to meet the requirements, submit a firm fixed-price quotation, and provide a Unique Entity ID. Award will be made on a lowest price technically acceptable basis. Quotes are due by September 5, 2024, and the government intends to award without discussions but reserves the right to conduct discussions.

STATEMENT OF WORK
BACKGROUND
USDA, Agricultural Research Service (ARS), Animal Genomics and Improvement Laboratory (AGIL) in Beltsville, MD request Enzymatic Methyl-Seq (including DNA extraction/QC, library preparation, and sequencing) service for 106 samples. The Lab collected 36 liver, 36 muscle, 22 skin, and 12 mammary gland tissue samples from cattle.
REQUIREMENTS AND DELIVERABLES
Genomic DNA will be extracted from liver, muscle, skin, and mammary gland samples. Firstly, a library will be made by ligating enzymatic methyl-seq adaptor to sheared end repaired/dA-tailed genomic DNA. This will be followed by two sets of enzymatic conversion steps to differentiate unmethylated cytosines from 5mC/5hmC. After PCR amplification of libraries, each library will be sequenced to generate at least 150 Gb EM-seq data using an Illumina PE150 platform to study genome-wide DNA methylation. These jobs will be performed in the service provider’s facility from September 12, to December 31, 2024. All data analysis will be performed by USDA AGIL.
ESTIMATED PERIOD OF PERFORMANCE
September 12, 2024 to December 31, 2024
