Statement of Work Cell Line Expressing Human Voltage-gated Sodium Channels
25 August 2022
RFQ # W9ZLK-22-Q-0062
1. PROJECT TITLE: Purchase of Cell Line Expressing Human Voltage-gated Sodium Channels.
This is a solution to measure the effects of synthesized conotoxins on its respective target.
2. BACKGROUND:
A cell line is described as a defined population of cells that can be maintained in culture for an extended period of time, retaining stability of certain phenotypes and functions. Utilizing a cell line allows for investigation of specific targets that are typically difficult to assess in vivo, such as ion channels. Many chemical warfare agents affect the structure and function of ion channels.
Growing cell lines that express specific ion channels allows DEVCOM CBC investigators to test a library of compounds and characterize the change in activity in a manner that was specifically designed for electrophysiological assays, such as the Sophion Q-Patch II Automated Patch Clamp system which is currently in use in the Threat Agent Sciences Division of R&T Directorate. This also reduces the number of animals needed to understand the effects of these agents on ion channel activity. DEVCOM CBC investigators can then compare potency and efficacy of chemical agents to benchmark compounds to rank and/or prioritize our chemical threat list.
3. SCOPE:
The scope of this contract is to purchase required cell lines for the associated project goal of measuring current through ion channels of individual cells when exposed to chemical agent. This will contribute to better risk assessments for chemical threat agents.
