About this file

This document is a Statement of Need (SON) for a federal contract opportunity issued by the National Oceanic and Atmospheric Administration (NOAA) to procure 750 hollow membrane filters for use in a salmon ocean behavior and distribution study. The filters are essential for extracting environmental DNA (eDNA) from water samples to identify the diverse range of organisms present, which is critical for understanding the complexity and health of marine ecosystems.

The SON provides the technical specifications for the required filters, including that they must be capable of filtering high volumes of water without compromising DNA integrity, have an optimal pore size for retaining a wide range of organismal DNA, be durable enough for repetitive DNA extraction, and be compatible with the metabarcoding machinery. NOAA intends to award a single-source, firm-fixed-price contract to RKS Laboratories for the filters, as detailed in the related federal contract opportunity. Responses to this notice of intent are due by Tuesday, June 25, 2024, and must include technical data, cost information, and convincing evidence of the supplier's capability to meet the requirement.

STATEMENT OF NEED

Requisition 143041-24-0156

PURPOSE:

NOAA/NMFS has studied salmon ocean distribution and survival vigorously since 1998. In that time, little has been learned about the functional dynamics between salmon and predators at the scale of fronts plumes – widely regarded as critical areas defining ecosystem functionality. The filters being requested for this purpose are specific to sampling eDNA around and on fronts in an effort to diagnose these interactions in California Current.

The Salmon Ocean Behavior and Distribution Study requires the procurement of 750 Hollow Membrane filters for a vital aspect of its research—the extraction of DNA from ocean water samples for metabarcoding analysis. These filters are instrumental in identifying the diverse range of organisms residing within the ocean's waters, which is a critical component in understanding the complexity and health of marine ecosystems. The metabarcoding process allows for the rapid assessment of biodiversity, enabling researchers to detect and monitor the presence of various species, including those that are elusive or endangered.

TECHNICAL SPECIFICATIONS:

The filters we are ordering from RKS Laboratories are specifically produced for the instruments we are using to analyze continuous eDNA sampling (750 hollow membrane filters, crafted specifically for the laboratory processing samples). The specified Hollow Membrane filters, essential for the DNA extraction machine, are designed to capture even the smallest particulate matter from water samples, ensuring a comprehensive DNA yield. In a metabarcoding context, after the eDNA has been extracted from the water samples using the filters, the DNA can then be amplified using PCR with specific primers that target the genetic regions of interest. The amplified DNA is then sequenced, often using next-generation sequencing platforms like the Illumina MiSeq, to identify the different species present in the sample.

For such sensitive processes, the precision and compatibility of the filters with the extraction kits and machinery are crucial for accurate DNA recovery and subsequent data analysis. The Hollow Membrane filters specified for this project would be selected for their pore size, material, and durability to ensure they are capable of withstanding the rigors of the extraction process while providing a high yield of quality DNA suitable for metabarcoding analysis.

Each filter must meet the following criteria:

· Be capable of filtering a high volume of water without compromising the integrity of the DNA.

· Have a pore size that is optimal for the retention of a wide range of organismal DNA.

· Be durable enough to maintain performance under the repetitive process of DNA extraction.

· Have compatibility with the metabarcoding machinery in terms of size and functional design.

REQUESTED DELIVERY DATE / PERIOD OF PERFORMANCE:

The delivery of the Hollow Membrane filters is requested as soon as possible, considering the project's timeline and the preparatory work required before the main metabarcoding analysis can begin.
