FAR 52.252-2 CLAUSES INCORPORATED BY REFERENCE

NON-COMPETITIVE COMBINED SYNOPSIS / SOLICITATION

Title: Request for a Bioruptor® Pico Sonication Device

(i) This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in FAR Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested, and a separate written solicitation will not be issued.

(ii) The solicitation number is 75N98027Q00035. This solicitation is issued as a Notice of Intent (NOI) for a non-competitive combined synopsis/solicitation to award a contract or purchase order without providing for full and open competition (including brand-name, if applicable). This acquisition is conducted under the authority of FAR 12.603, Streamlined Solicitation for Commercial Products or Commercial Services, and is not expected to exceed the simplified acquisition threshold. Additional authority includes 41 U.S.C. 1901 and FAR 6.302-3(a)(2)(ii).

The National Institutes of Health (NIH), Office of the Director, Office of Acquisitions, intends to negotiate and award a firm-fixed-price purchase order on a non-competitive basis to Hologic Sales and Services, LLC (Hologic Diagenode), 400 Morris Ave, Suite 101, Denville, NJ 07834 for one (1) Bioruptor® Pico Sonication System and associated starter maintenance/consumable accessories.

Justification for the Award: The National Heart, Lung, and Blood Institute (NHLBI) Systems Biology Center requires the Bioruptor® Pico Sonication System to maintain strict continuity and experimental consistency across ongoing chromatin immunoprecipitation coupled with next-generation sequencing (ChIP-seq) and related epigenomic assays. The laboratory previously established its foundational ChIP-seq protocols and baseline datasets using the Bioruptor® Pico platform. Prior testing with single-probe sonicators and single-sample focused acoustic systems failed to maintain consistent chromatin fragment sizes due to single-sample processing limitations and probe decay. The Bioruptor® Pico uniquely combines cooled water-bath indirect sonication with simultaneous multi-sample processing in Pico-specific 0.65 mL (up to 12 samples) and 1.5 mL (up to 6 samples) microtubes. Transitioning to a different acoustic platform or sample vessel would alter shearing kinetics, fragment-size distributions, chromatin recovery, and ChIP enrichment, invalidating comparison with prior datasets and imposing an unacceptable burden in staff time, reagent costs, and loss of scarce primary and rare cell biological material. Hologic Sales and Services, LLC (Hologic Diagenode) is the original equipment manufacturer and exclusive distributor of the Bioruptor® Pico system, with no authorized third-party resellers.

(iii) The solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2026-01, effective 03/13/2026.

(iv) The associated NAICS code is 334516 (Analytical Laboratory Instrument Manufacturing), and the small business size standard is 1,000 employees. This requirement is not set aside for small business.

(v) Description of Requirement:

The NHLBI Systems Biology Center requires one (1) Bioruptor® Pico Sonication System and associated starter maintenance and consumable accessories to prepare chromatin for ChIP-seq and related epigenomic studies of transcriptional regulation, chromatin looping, enhancer activity, and epigenetic control of gene expression in primary cells and rare cell populations. Consistent chromatin fragmentation into uniform DNA–protein complexes directly impacts data quality, resolution, reproducibility, and the ability to accurately identify regulatory elements and protein–DNA interactions across the genome.

Minimum Functional and Performance Requirements (Salient Characteristics):

• Chromatin Shearing: Must reproducibly fragment chromatin from primary cells and rare cell populations into uniform DNA–protein complexes suitable for ChIP-seq while preserving epitopes required for immunoprecipitation.

• Control and Reproducibility: Must provide adjustable sonication ON/OFF durations and cycle counts so that sample-specific shearing conditions can be optimized and repeated across experiments.

• Sample Protection: Must provide indirect sonication in closed tubes and an integrated cooling system to control sample heating during processing and reduce the risk of cross-contamination and sample damage.

• Low-Input Processing and Tube Compatibility: Must process low-input samples in compatible 0.65 mL and 1.5 mL microtubes, with holders that support consistent tube positioning and parallel simultaneous processing of multiple samples (12 samples in 0.65 mL tubes or 6 samples in 1.5 mL tubes per run).

• Maintenance, Installation, and Support: Must include the Essential Maintenance Starter Pack providing one (1) year of comprehensive support covering manufacturing repairs and firmware updates, a quality control kit including a high-quality tube holder, a bonus pack of shearing consumables, and no-charge installation and training.
