About this file
This is a Request for Quotation (RFQ) and Public Notice of Proposed Acquisition issued by the National Institutes of Health (NIH), National Heart, Lung, and Blood Institute for Cryo-Electron Microscopy (Cryo-EM) Services.
The National Institute on Deafness and Other Communication Disorders (NIDCD), Laboratory of Cell Structure and Dynamics requires specialized cryogenic electron microscopy services to prepare cellular specimens and perform in situ cryo-electron tomography of hair cell structures. The contractor must provide an integrated cryogenic workflow including Correlative Light and Electron Microscopy (CLEM), fluorescence-guided identification of regions of interest, cryogenic focused ion beam (cryo-FIB) milling, specimen preparation, cryogenic screening, high-resolution cryo-ET data acquisition, and initial data preprocessing and tomographic reconstruction. Key contractor responsibilities include providing access to all necessary instrumentation and software, qualified technical personnel experienced in cryo-EM and cryo-FIB techniques, technical assistance for optimization of imaging parameters, maintenance of appropriate cryogenic conditions, data delivery in agreed electronic formats, and support for direct Government scientist participation in specimen screening and data acquisition activities. The workflow must enable real-time scientific decision-making and continuity among preparation, targeting, screening, and acquisition phases.
The RFQ is not a small business set-aside and is being conducted under FAR Part 13 Simplified Acquisition Procedures as a single-sole source, non-competitive action. The University of Virginia Molecular Electron Microscopy Core was identified through market research as the only source capable of meeting the complete technical and operational requirements. Quotations must be received by September 16, 2026 at 2:00 PM EST, with proposed award dates of September 30, 2026 through September 29, 2027. Responses must include unit pricing, list price, shipping and handling costs, delivery period, prompt payment discount terms, F.O.B. point, DUNS number, Taxpayer Identification Number, and business size certification, with all offerors requiring active SAM registration. Contact: Roxanne Burkett (Roxane.burkett@nih.gov) and Andrew Dinh (Andrew.dinh@nih.gov).

14.Description: The Laboratory of Cell Structure and Dynamics (LCSD), NIDCD, requires specialized cryogenic electron microscopy services to prepare cellular specimens and perform in situ cryo-electron tomography (cryo-ET) of hair cell structures.
The objective of this requirement is to provide an integrated cryogenic workflow that enables Correlative Light and Electron Microscopy (CLEM), fluorescence-guided identification of biologically relevant regions of interest, cryogenic focused ion beam (cryo-FIB) milling and preparation of electron-transparent lamellae, cryogenic specimen screening, high-resolution cryo-ET data acquisition, and initial preprocessing and tomographic reconstruction.
The Contractor shall:
1. Provide access to all cryogenic microscopy instrumentation, software, computational resources, and supporting infrastructure necessary to perform the services described in this SOW.
2. Provide qualified technical personnel with experience in cryogenic electron microscopy, cryo-FIB specimen preparation, correlative fluorescence/electron microscopy, and cryo-electron tomography.
3. Coordinate fluorescence-guided targeting, cryo-FIB lamella preparation, specimen screening, cryogenic imaging, and tomography acquisition to minimize unnecessary specimen handling and maintain appropriate cryogenic conditions.
4. Perform or support site-specific preparation of electron-transparent cryogenic lamellae at identified regions of interest.
5. Provide technical assistance for optimization of specimen targeting, milling, imaging, and tomography acquisition parameters as appropriate for the biological specimens.
6. Perform cryogenic specimen screening and high-resolution cryo-ET data acquisition using appropriate low-dose methodologies.
7. Provide current acquisition and automation software necessary for efficient and reproducible tilt-series data collection.
8. Provide initial data preprocessing and tomographic reconstruction sufficient to evaluate data quality and experimental success.
9. Maintain appropriate cryogenic conditions throughout specimen preparation, transfer, screening, and imaging to minimize contamination, devitrification, specimen damage, or loss.
10. Provide generated experimental data and associated reconstruction outputs to the Government in an agreed-upon electronic format.
11. Provide technical staff assistance necessary to support completion of the required experimental activities.
12. The facility shall support an integrated workflow in which fluorescence localization, cryo-FIB preparation, cryogenic screening, and cryo-ET acquisition can be coordinated with minimal unnecessary specimen transfer or disruption.
13. The facility shall permit appropriately qualified and trained Government scientific personnel to participate directly in specimen screening, identification of regions of interest, targeting, and data-acquisition activities, subject to applicable facility safety, training, and instrument-authorization requirements.
14. The workflow shall permit real-time or near-real-time scientific decision-making during specimen targeting and imaging so that biological expertise can be applied to selection of regions of interest.
15. The Contractor shall provide technical personnel capable of working collaboratively with Government scientists to optimize cryogenic specimen preparation, targeting, and imaging procedures for specialized biological specimens.
16. The facility shall provide sufficient continuity among specimen preparation, targeting, screening, and cryo-ET acquisition to support reproducible application of established experimental methodologies.
15. Statutory Authority:
This acquisition is conducted under the authority of the Federal Acquisition Regulation (FAR) Part 13—Simplified Acquisition Procedures, Subpart 13.106-1
(b) (1), Soliciting from a single source and is not expected to exceed the simplified acquisition threshold. Contracts awarded using FAR Part 13— Simplified Acquisition Procedures are exempt from the requirements of FAR Part 6—Competition Requirements.
16. Single-sole Source (Including brand-name) Determination:
The determination by the Government to make a single-sole source (Including brand-name) award is based upon the market research conducted as prescribed in FAR Part 10—Market Research, specifically the results of the market research concluded: The University of Virginia (UVA) Molecular Electron Microscopy Core is the only identified source capable of meeting the complete technical and operational requirements of this acquisition without substantial duplication of workflow-development effort, additional specimen risk, and research delay. No alternative source identified through the market research provides the complete combination of capabilities, access, specimen-specific workflow, and established expertise required by LCSD in an immediate operational form.
17. Closing Statement:
This notice is not a request for competitive proposals. However, interested parties may identify their interest and capability to respond to this notice.
Responses to this notice shall contain sufficient information to establish the interested parties’ bona-fide capabilities for fulfilling the requirement and include:
unit price, list price, shipping and handling costs, the delivery period after contract award, the prompt payment discount terms, the F.O.B. Point (Destination or Origin), the Dun & Bradstreet Number (DUNS), the Taxpayer Identification Number (TIN), and the certification of business size. All offerors must have an active registration in the System for Award Management (SAM) www.sam.gov.”
A determination by the Government not to compete this proposed action based upon responses to this notice is solely within the discretion of the Government.
The information received will normally be considered solely for the purposes of determining whether to proceed on a non-competitive basis or to conduct a competitive procurement.
All responses must be received by September 16, 2026 at 2:00PM EST.
Responses must be submitted via email to the Contract Specialist and Contracting Officer identified above.
“All responsible sources may submit a bid, proposal, or quotation which shall be considered by the agency.”
