Specific work includes monitoring plasma concentrations of the parent drug and 1-MNA (1-methyl nicotinamide) at predetermined time points (0, 5, 15, 30 minutes, 1, 2, 4, 8, 12, and 24 hours post-dose) with n=3 mice per time point, and delivering concentration versus time curves and PK parameters in an Excel report. The estimated level of effort is 1 PhD-level manager and 1 PhD-level scientist for 3 months total. The period of performance is September 4, 2026 to March 3, 2027, with documentation and results to be delivered to NCATS within three months of purchase order receipt via email to Samarjit Patnaik (samarjit.patnaik@nih.gov) and Vinoth Chenniappan (vinothkumar.chenniappan@nih.gov). The Government will provide the three NNMT inhibitor compounds. Evaluation factors include price, technical capability, past performance, and delivery schedule. All responses must include unit and total price, shipping and handling costs, delivery timeframe, delivery terms, F.O.B. point, product description, Unique Entity Identifier (UEI), Taxpayer Identification Number (TIN), and business size certification. STATEMENT OF WORK (SERVICES)
(SOW)
GENERAL INFORMATION
Title of Project:
Developing selective small-molecule inhibitors of NNMT for chronic kidney disease
Statement of Need and Purpose:
This purchase order supports pharmacokinetic (PK) and pharmacodynamic (PD) studies of three NNMT inhibitors in mice following oral (PO) and IV (intravenous) administration at predecided single dose.
Samples will be collected from plasma at the following time points: 0 min, 5 min, 15 min, 30 min, 1 h, 2 h, 4 h, 8 h, 12 h, and 24 h (n=3 mice per time point). Drug and 1-MNA (1-methyl nicotinamide) concentrations will be measured in plasma evaluate systemic exposure 1-MNA suppression, and facilitate advancement of compounds into preclinical development.
Background Information and Objective:
The National Institutes of Health (NIH) is the nation’s leading medical research agency and the primary Federal agency whose mission is to seek fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability, conducting, supporting and making medical discoveries that improve people’s health and save lives.
The National Center for Advancing Translational Sciences (NCATS, Early Translation Branch) is a part of the National Institutes of Health (NIH), whose mission it is to catalyze the generation of innovative methods and technologies that will enhance the development, testing and implementation of diagnostics and therapeutics across a wide range of human diseases and conditions.
NNMT is a methyl transferase implicated in multiple metabolic diseases with a lead inhibitor from Astra Zeneca in Phase 1 clinical trials for chronic kidney disease (CKD) in diabetes patients. NCATS in collaboration with U Chicago has developed orally bioavalble NNMT inhibitors for cancer therapy and is engaged in a camaoign to develop NNNMT inbhitors with longer half life than the current lead 960. These PK/PD studies will evaluate lead compounds and compare them with the current lead 960.
Period of Performance:
Documentation of results and presentation of these results to NCATS via email with the next three months after receipt of PO.
