Version 12-23-2025-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION:

NCATS DPI Functional Genomics Lab (FGL) needs one Agilent 4200 TapeStation instrument with laptop to check size, amount and integrity of DNA and RNA samples

1. Statement of Need and Purpose: FGL needs one Agilent 4200 TapeStation instrument with a laptop computer to check the size, amount, and integrity of deoxyribonucleic acid and ribonucleic acid samples before they are sent for sequencing. This capability is needed to support perturbation screening studies in more complex biological models and to ensure that only high-quality samples move forward, which helps avoid failed sequencing runs, delays, and unnecessary cost. The requested system must be a benchtop, automated sample quality assessment instrument that can process up to 96 samples in a single run, produce results in less than 90 minutes for a full 96-well plate, and support a broad range of deoxyribonucleic acid and ribonucleic acid applications using prefilled ScreenTape devices.

2. Background Information and Project details: This equipment is replacing an older instrument, TapeStation 2200, whose support ended on December 31, 2022, and which is not compatible with NGS library QC. This acquisition would allow us to perform the complete workflow of the single cell sequencing based CRISPR screens up until the NGS library QC, which opens up new research capabilities within functional genomics screens. Since the last step of this workflow is a sequencing step that we usually send out to commercial entities since we do not have access to a sequencer, being able to QC the NGS libraries on a compatible TapeStation prevents the samples from being rejected by the sequencing facility, preventing significant delays. All FGL projects are under 1 ZIC TR000041-10 and this would be the acquisition of a TapeStation 4200 to be able to QC the DNA being sent out for sequencing downstream of PerturbSeq screens that we would be performing in collaboration with the intramural NIH community to understand the effects of gene perturbations in complex models at the single cell level

3. Generic Name of Product: an automated electrophoresis solution for sample quality control (QC) that can assess DNA and RNA sample size, quantity, and integrity.

4. Purchase Description and Salient Characteristics: G2991BA Walk-away automation in a benchtop system for the analysis of up to 96 DNA or RNA samples Consistent cost-per-sample Fast results, capable of analyzing a full 96-well plate in less than 90 minutes Prefilled Screen Tape devices Broad DNA and RNA application portfolio
