STATEMENT OF WORK (SERVICES)
(SOW)
GENERAL INFORMATION
Title of Project:
Synthesis of compounds and intermediates for Ewing sarcoma project
Statement of Need and Purpose:
This purchase order supports the synthesis of compounds and intermediates for a project that is developing compounds targeted towards Ewing sarcoma. The custom non-GMP synthesis, purification, analytical characterization, and delivery of 5 grams each of these small-molecule compounds will be prepared for research purposes in support of ongoing NCATS medicinal chemistry and preclinical development activities.
The purpose of this effort is to obtain research-grade material of each target compound at a minimum purity suitable for further scientific evaluation.
Background Information and Objective:
Scientists from NCATS and the University of North Carolina are working together to find a new treatment for Ewing Sarcoma, a rare bone and tissue cancer, by fixing how DNA is packaged inside cancer cells. In healthy cells, DNA is wrapped up like a neatly organized library where only specific genes are "open" and readable, but in Ewing Sarcoma cells, this system breaks down, leaving the wrong parts of the DNA accessible and causing the cells to become cancerous. To map out exactly where these errors are, the researchers use a specialized scanning tool called ATAC-Seq, which identifies which parts of the DNA are open or closed.
The team is seeking to purchase and test this specific list of chemical compounds to see if they can act as a reset button, forcing the chaotic DNA structure of the cancer cells to look and behave like normal, healthy cells again. Ultimately, their goal is to prove that fixing this DNA packaging will stop the cells from being cancerous, either by using these exact compounds or developing new medicines based on them.
To support these research activities, the Contractor shall synthesize and deliver the five compounds listed below under non-GMP conditions, with a target purity of greater than or equal to 95%, together with a Certificate of Analysis (COA) for each compound.
Target Compounds:
1. N-Methyl-N-(3-(((2-((2-oxoindolin-5-yl)amino)-5-(trifluoromethyl)pyrimidin-4-yl)amino)methyl)pyridin-2-yl)methanesulfonamide ABCL Code: ABCL-0482 Quantity: 5 g
