Version 12-23-2025-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

1. Statement of Need and Purpose: The purpose of this acquisition is to obtain one (1) QIAsprint Connect System to support high-throughput nucleic acid extraction and purification for ongoing stem cell research activities. Current and planned projects require the processing of large numbers of biological samples for downstream molecular analyses, including quantitative PCR, transcriptomic, genomic, and epigenomic studies.

The requested system will automate the extraction of DNA and RNA from multiple sample types and enable parallel processing of up to 192 samples in a single run. This capability will increase laboratory throughput, reduce hands-on processing time, minimize the risk of sample handling errors and contamination, and improve consistency across experiments. The resulting nucleic acids will be used to characterize stem cell-derived products, evaluate differentiation processes, and support studies investigating disease mechanisms and cellular development.

Acquisition of this system is necessary to maintain project timelines, support expanding research demands, and provide reliable sample preparation for current and future studies.

2. Background Information and Project details: The Stem Cell Toxicology Laboratory (SCTL) conducts research focused on generating human pluripotent stem cell (hPSC)-derived cell types that support New Approach Methodologies (NAMs) and other human-relevant in vitro models. These efforts contribute to the development and evaluation of alternatives to traditional animal testing and support regulatory science initiatives.

Characterization and validation of hPSC-derived cell products require routine analysis of gene expression and other molecular endpoints throughout the differentiation process.

These analyses depend on the efficient extraction of high-quality nucleic acids from large numbers of samples. The extracted DNA and RNA are used in quantitative PCR assays and multi-omics applications to assess cellular identity, differentiation status, and biological function. Resulting data help identify genes and pathways involved in cell development, lineage specification, and disease-related processes.

The increasing scale of these studies necessitates an automated, high-throughput extraction platform capable of generating reproducible results while reducing labor-intensive manual processing.
