Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

1. Statement of Need and Purpose: The purpose of this acquisition is to purchase long read sequencing kits to support the creation of a single cell atlas from human brain tissue that is broadly useful to the neurodegenerative disease research community. This atlas aims to provide integrated genetic, epigenomic, and transcriptomic data in order to facilitate the rapid translation from genetic variant to functional consequence. The execution of this work is centered on three underlying principles. 1) Leveraging samples with existing complementary data. 2) Introducing ancestral diversity in the experiment. 3) Making data accessible and actionable for the research community

2. Background Information and Objective:

The Brain Atlas project has made progress on multiomic analysis (gene expression and chromatin accessibility) of 350 healthy control samples from the prefrontal cortex (PFC).

This includes identifying eQTLs, and ongoing work on caQTL and gene regulatory analyses. Preliminary results also exist for substantia nigra (SN) samples, showing decreased dopaminergic neurons in cases versus controls. An analysis pipeline has been developed for PFC data that will be used for SN and hippocampus (HP) samples.
