About this file

This Request for Quotation (RFQ) seeks two Thermo Scientific Exploris 120 Orbitrap Mass Spectrometry and Ultra-High Performance Liquid Chromatography (UHPLC) systems for the U.S. Food and Drug Administration's Office of Pharmaceutical Quality Research in St. Louis. The procurement is a small business set-aside under NAICS code 334516, with a size standard of 1,000 employees. The systems will support testing and research on nitrosamine impurities in drug products, leveraging the instruments' ability to analyze compounds with a mass range of 40-3000 m/z and provide high-resolution, accurate mass measurements.

The contract includes delivery, installation, qualification, and instrument familiarization, with a base performance period of approximately 17 months. This includes 90 days for initial delivery and installation, instrument training, and a one-year initial warranty. An optional 12-month preventative maintenance period follows the warranty. The FDA specifically notes that these Exploris 120 systems, at half the cost of the Exploris 480 model, will meet their testing requirements for analyzing small nitrosamines and drug substance related impurities in prescription and over-the-counter drug products. The solicitation is issued directly by the Department of Health and Human Services Food and Drug Administration Office of Acquisition and Grant Services.

Section 3 – Statement of Work

Thermo Scientific Exploris 120 Orbitrap MS and UHPLC Systems Background

The Office of Pharmaceutical Quality Research – St. Louis (OPQR – St. Louis) has been providing laboratory testing and research support and has been working with the Office of Pharmaceutical Quality (OPQ) Nitrosamine Taskforce since the beginning of the nitrosamine contamination issue which started in early 2018. Six years later and the nitrosamine issue has not gone away but has gotten bigger in scope. The problem started with small nitrosamine contamination (i.e. NDMA, NDEA, etc.) in angiotensin receptor blocker (ARB) drug products. However, the contamination issue over the years has grown to include nitroso drug substance related impurities (NDSRIs) and fragment NDSRIs in both prescription and over the counter (OTC) drug products.

OPQR – St. Louis requires two new Exploris 120 Orbitrap MS systems, or equal, to support testing and research on nitrosamine impurities in drug products and substances. The Exploris 120 Orbitrap MS system can analyze compounds with a wide variety of sizes from 40 – 3000 m/z. The excellent sensitivity and resolution of this mass spectrometer will be used to identify nitrosamine drug substance related impurities (NDSRIs) by matching accurate mass measurements with calculated monoisotopic masses, and to develop fit-for-purpose methods to accurately detect and quantitate NDSRIs. Reference standards for these NDSRIs are not readily available and take on average 8 to 10 weeks to custom synthesize. Therefore, surveillance testing of drug products suspected to contain NDSRIs would take a lot of time to perform and complete. Two systems are required due to the high number of high priority nitrosamine projects and tight timelines.

The benefit of the lower range of this instrument is that it will also enable the analysis of small molecule nitrosamines (e.g., NDMA & NDEA) and other drug product and drug substance impurities such as nitrite and dimethylamine with great accuracy and sensitivity. Traditionally, these impurities are analyzed using Ion Chromatography which does not provide sufficient sensitivity, precision, or speed of analysis for these common impurities.

Xcalibur software and Chromeleon Chromatography Data System (CDS) that the Thermo Scientific Exploris 120 and Vanquish UHPLC system utilize are already in the approved MAT list since three other Exploris 480 systems are already in service at OPQR – St. Louis’ Precision Analytics Mass Spectrometry Laboratory as of 2023.

Objective

To purchase two Thermo Scientific Exploris 120 Orbitrap MS and UHPLC systems to enhance CDER/OPQ/OPQR – St. Louis’ capability in characterizing drug substances and drug products with potential nitrosamine impurities in support of domestic drug manufacturing. The three previous Thermo Scientific Exploris Orbitrap MS systems that were purchased by OPQR – St. Louis have been the top of the line model which is the Exploris 480. However, the purchase of the base model, Exploris 120, would be more than adequate and cost effective but still meet the testing requirements necessary to test for small nitrosamines, nitroso drug substance related impurities (NDSRIs) and fragment NDSRIs accurately and precisely in prescription and over the counter (OTC) drug products at half the cost of an Exploris 480 MS and UHPLC system.

Scope

This statement of work covers the delivery, installation, qualification, and instrument familiarization on two new Exploris 120 Orbitrap MS and UHPLC systems or equal. Upon installation, operation of the instrument and performance qualification shall be demonstrated.

Detailed Technical Requirements

The proposed acquisition package should include the high resolution and accurate mass (HRAM) spectrometers, ultra-high performance liquid chromatograph (UHPLC), IQ/OQ of both LC-MS systems, computers with preloaded Thermo Scientific Chromeleon chromatography data system, and vendor provided instrument and software familiarization. The mass spectrometer system should also include a power conditioner (P/N C0012-01-00080) and a power distribution unit module (P/N C0202-99-00182). Three-day onsite training is not requested since OPQR analysts are already using three other orbitrap Exploris 480 LC-MS systems.

Once the Government determines the quoter that is the best-suited (i.e., the apparent successful quoter), the Government reserves the right to communicate with only that quoter to address any remaining issues, if necessary, and finalize a contract or order with that quoter. These issues may include technical and price. If the parties cannot successfully address any remaining issues, as determined pertinent at the sole discretion of the Government, the Government reserves the right to communicate with the next best-suited quoter based on the original analysis and address any remaining issues. Once the Government has begun communications with the next best-suited quoter, no further communications with the previous quoter will be entertained until after the contract or order has been awarded. This process shall continue until an agreement is successfully reached and a contract or order is awarded.
