INTRODUCTION
THIS IS A PRE-SOLICITATION NON-COMPETITIVE (NOTICE OF INTENT) SYNOPSIS TO AWARD A CONTRACT
OR PURCHASE ORDER WITHOUT PROVIDING FOR FULL OR OPEN COMPETITION.
The National Institutes of Health, National Institute of Child Health and Development (NICHD), in support of the National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK), intends to award a purchase order without providing for full and open competition to METABOLON, INC. for Metabolomic Analysis Services.
BACKGROUND
The NIH, NIDDK, Liver Diseases Branch, Immunology Section performs basic and translational research to understand (1) innate and adaptive immune responses to hepatitis viruses and (2) the regulation of the hepatic and systemic immune responses by the microbiome. The section is studying the effect of microbiomes on host physiology and immune responses. To this end, investigators in the section have found that the gastrointestinal microbiota affect energy expenditure via an effect on the thermogenic adipose tissue. The objective of this procurement is to acquire services needed to identify the causal agents for the Liver Diseases Branch. Specifically, to analyze metabolites in 24 brown and 24 white adipose tissue samples and 24 plasma samples of two groups of mice that differ in their microbiota and have shown differential weight gain and metabolism upon high-fat diet.
JUSTIFICATION
To our knowledge, only Metabolon has the ability to detect and identify small molecule metabolites of over 60 different classes based on ultrahigh performance liquid chromatography/tandem accurate mass spectrometry (UHLPC/MS/MS) methods with peak identification, deconvolution and comparison to a reference library of more than 4000 known metabolites. More importantly, Metabolon previously analyzed baseline samples for NIDDK from the same mice as this order will use, therefore the Liver Diseases Branch needs to send the current samples to the same vendor so that comparison of the results can match the protocols used previously. A switch now to alternative services would require new baseline samples to start from, causing delays and having impacts on institute resources. The Global Discovery Panel - Data + Interpretation Metabolomic Analysis Service is proprietary to Metabolon Inc, therefore they are the only source capable of providing it.

REGULATORY AUTHORITY
The resultant contract will include all applicable provisions and clauses in effect through the Federal Acquisition Regulation (FAR) Circular (FAC) 2025-05 effective 08/07/2025. This acquisition is conducted under the procedures as prescribed in FAR subpart 13-Simplified Acquisition Procedures at an amount not exceeding the simplified acquisition threshold ($250,000) and FAR Subpart 12- Acquisition of Commercial Items.

DESCRIPTION OF REQUIREMENT
The Liver Diseases Branch requires identification of metabolites that are differentially expressed in brown and white adipose tissue of mice that are protected from high-fat diet-induced obesity and that are not protected.
The vendor shall: Combine multiple different ultra-high performance liquid chromatography/tandem accurate mass spectrometry (UHPLC/MS/MS) methods with peak identification, deconvolation and chemical intelligence. Identify more than 5000 metabolites via comparison to a reference library of metabolites.
Identify endogenous metabolites and those derived from microbiota and food. Provide both data analysis by the vendor and a portal where NIDDK can further analyze data and identify pathways of interest.
Task Area 1 – Analyze and identify metabolites in 24 brown and 24 white adipose tissue samples and 24 plasma samples and provide relative concentrations in excel table format. Provide statistical analysis, and summary and interpretation report.
Task Area 2 –Provide access to Client Portal with data analysis and visualization tools.
Task Area 3 – Provide spectral data in mzML format.
Period of Performance: 5 months from date of award. The vendor shall deliver results within 16 weeks after the date that all biological samples are received by the vendor in acceptable condition.
CLOSING STATEMENT
This synopsis is not a request for competitive proposals. However, interested parties may identify their interest and capability in response to this notice and may submit a capability statement, which shall be considered by the agency. Responses to this notice shall contain sufficient information to establish the interested parties’ bona-fide capabilities for fulfilling the requirement. A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government. The information received will normally be considered solely for the purposes of determining whether to proceed on a non-competitive basis or to conduct a competitive procurement.
All respondents must have an active registration in the System for Award Management (SAM) www.sam.gov. All responses to this notice shall be submitted electronically by 12:00 PM Eastern Daylight Time, on September, 17th, 2025 to jacob.balenson@nih.gov and shall include pre-solicitation # 75N94025Q00189 in the subject line.
