Salient Characteristics:

STED and Confocal Core microscope provides a modular super-resolution imaging platform that combines confocal imaging, 2D/3D STED capability, tunable spectral detection, adaptive illumination, and integrated analysis software in a configuration suited for high-resolution fixed-sample and live-cell imaging in a shared research environment.

• Integrated confocal and STED core microscope with modular expansion Configured as a working confocal and STED microscope from the outset, while also serving as the base platform for additional modules in the future. This provides immediate super-resolution capability now without limiting future expansion as research needs evolve.

• Motorized XY stage and piezo Z stage for precise 3D and multi-position imaging includes a fully software-integrated motorized XY stage with a universal holder for plates, dishes, and slides, plus a 200 µm piezo z-stage for precise and rapid z-stacks. This is distinctive because it supports a wide range of sample formats and enables automated tiling, repeated positioning, and fast volumetric imaging with the precision needed for super-resolution datasets.

• QUADScan beam scanning for large images and fast acquisition is patent-protected QUADScan technology with field number 11.3, image sizes up to 16,000 × 16,000 pixels, and point-scanning rates up to 2.6 kHz with 4.2 frames per second at 512 × 512. This is a notable advantage because it supports both very large, information-rich images and fast data collection on the same platform, making the system suitable for detailed structural work as well as higher-throughput acquisition.

• High-power 775 nm STED laser and the easy3D module, which uses spatial light modulator technology for seamless control of light distribution in both 2D and 3D STED. The performance is below 30 × 30 nm in 2D and below 100 × 100 × 100 nm in 3D, with typical values of 25 × 25 nm and 80 × 80 × 90 nm, respectively. This enables nanoscale imaging beyond the reach of conventional confocal microscopes and enables both planar and volumetric super-resolution on a single platform.

• FLEXPOSURE adaptive illumination for live-cell compatibility and reduced bleaching.

The FLEXPOSITION package adaptively adjusts STED power on a pixel-by-pixel basis to minimize light dose while maintaining maximal resolution, achieving light-dose reductions of up to two orders of magnitude, reduced bleaching, and enhanced signal. It makes high-resolution STED more practical for sensitive specimens and extended imaging sessions, where photobleaching and phototoxicity are usually limiting factors.

• RAYSHAPE uses adaptive optics with a deformable mirror to dynamically correct specimen-induced aberrations on the fly as imaging depth changes. This preserves brightness and resolution over about a 200 µm z-range, uses 140 digitally controlled actuators that adjust within milliseconds, and improves imaging in thick, turbid, or refractive-index-mismatched samples compared with conventional correction-collar approaches that optimize only a limited depth range. This capability would strengthen deep-tissue confocal and 3D-STED imaging by maintaining a stable focus and sharper images throughout the specimen.

• MATRIX detector upgrade enables dramatic reduction of background levels. This technology combines pixel reassignment with an intelligent array deconvolution algorithm to enhance resolution.

• LiGHTBOX provides intuitive experiment planning, automated settings, rapid switching between parameter sets, time-lapse, frame and line stepping, multi-position acquisition, tiling and stitching, annotation, quantification, 3D rendering, and quick export of publication-ready figures.

This is distinctive because it lowers the training barrier for occasional users while still preserving advanced control for expert users, which is especially important for a shared institutional instrument.
