DNA Sequencing Equipment & Consumables Package

1.0 Introduction/Background

This Performance Work Statement (PWS) outlines the detailed requirements for a complete and integrated ensemble of laboratory test equipment, ancillary hardware, and chemical consumables. The ensemble is intended for conducting comprehensive DNA marking detection, QC authentication, PCR amplification, and capillary electrophoresis.

2.0 Scope

The contractor shall provide a turnkey laboratory equipment ensemble. The configuration must match or exceed the capabilities of the 3500 Genetic Analyzer 8 Channel (Model No: 622-0010) and the Applied Biosystems 2720 Thermal Cycler (Model No: 4359659R), paired with GeneMapper 6 software on a Windows 11 workstation. Additionally, a comprehensive starter kit of PCR reagents and swabbing tools is required. The contractor shall also provide 1 day of on-site training for up to 6 individuals upon installation.

3.0 Operational & Process Requirements

The equipment and consumables must support the following laboratory workflows:

• Sample Swabbing & Initial Detection: Utilization of UV Flashlights and IR Lasers to detect rapid reporters.

Swabbing of substrates (Nickel, Gold, Epoxy, Alumina) using Synthetic Nylon Swabs, Puritan Cotton Swabs, cut via razors into 0.2 mL Flat cap PCR reaction tubes.

• PCR Master Mix Preparation: Must be performed in a dedicated DNA Free Hood to prevent contamination.

• DNA Amplification: Utilizing the Thermal Cycler to perform precise heating and cooling cycles, incorporating fluorescent tags to the target DNA.

• Capillary Electrophoresis: Utilizing the 3500 Genetic Analyzer to separate DNA based on size, with software interpretation of specific 3-clone DNA combinations.
